Objective To investigate the effect of the eye-acupuncture therapy on hypothalamus and colonic substance P ( SP) and NK-1 receptors contents in rats with diarrhea-predominanirritable bowel syndrome ( D-IBS) and explore its related mechanism.Methods Forty male Wistar rats were equally randomized into control, IBS model, eye-acupuncture and acupuncture groups.IBS model was estab-lished by giving the rat with chronic stress stimulation ( cold-water swimming, tail clamping, electrical shock, etc.) for 18 days.Eye-acu-puncture of Xiajiao ( Low Energizer) Area, Pi ( Spleen) Area, Gan ( Liver) Area and Dachang ( Large Intestine) Area were given to the rat for 20 min, twice daily for 7 d.The expressions of SP mRNA and NK-1 mRNA were measured by RT-PCR, and the expressions of SP and NK-1 protein were detected by immunohistochemistry.Results Compared with the model group, the expressions of SP mRNA,NK-1 mRNA and SP, NK-1 protein in rat hypothalamus and colon after the eye acupuncture therapy were both obviously decreased (P<0.01).Conclu-sions Eye-acupuncture could reduce hypothalamus and colonic SP and NK-1 contents in D-IBS rats, which might play a role in relieving IBS in eye-acupuncture clinic.%目的:观察比较眼针及体针对腹泻型肠易激综合征(D-IBS)大鼠下丘脑及结肠组织P物质(SP)及神经激肽Ⅰ型受体(NK-1)表达的影响。方法将40只雄性SD大鼠随机分为正常组、模型组、眼针组、体针组4组,每组10只。采用慢性应激结合束缚方法建立IBS大鼠模型。眼针组取下焦区、大肠区、肝区、脾区进行针刺,治疗7 d。用免疫组化方法检测大鼠下丘脑及结肠组织中SP和NK-1蛋白的表达;用RT-PCR方法检测大鼠下丘脑及结肠组织中SP mRNA的表达。结果与模型组比较,眼针组大鼠下丘脑和结肠组织 SP和 NK-1蛋白的表达以及 SP mRNA的表达均明显减少(P<0.01)。结论眼针疗法优于体针,对D-IBS有治疗作用。
