长寿家系的白细胞端粒长度

         

摘要

Objective To explore the leukocyte telomere length ( LTL) and its possible relationship with human aging/longevity in a family-based cohort .Methods Relative telomere length of leukocytes was determined by real-time PCR for Bama long-lived families ( BLF, n=1 301) and was compared with that of local general population , Bama non-long-lived families (BNLF, n=283), and with long-lived fam-ilies and non-long-lived families from Pingguo (PLF, n=308; PNLF,n=482 respectively), a county which was about 200 km away from Bama area.All participants were Zhuang ethnically .Results (1) An inverse relationship was found between LTL and age independent of sex in every genealogy and all cohorts studied .(2)No significant difference on LTL was found between males and females in all age groups in BLF.Similar tendency was shown in most but not all age groups in the other families .(3)The LTL of the 90~year age group in BLF was longer than that of the PLF(1.11 vs 0.93,P=0.000)regardless of sex, indicating some beneficial mechanism in the maintenance of LTL . (4) The LTL of the 65~years old group in BLF ( corresponding to the first offspring of the long-lived individual ) was dramatically longer than that of the other families (BNLF, 1.24 vs 1.05, P=0.000.PLF, 1.24 vs 1.08, P=0.011.PNLF, 1.24 vs 1.13, P=0.021).In addition, the LTL of the oldest old (≥90 yrs) was similar with that of the 65~years old in the three control families , indicating that the off-spring of long-lived individuals might had inherited longer LTL from their parental generation .Conclusions Bama long-lived families present longer LTL with higher hereditary tendency which might be in part interpret the longevity phenomenon in the region .%目的:探讨长寿家系血液白细胞的端粒长度与人类衰老/长寿的关系。方法采用 real-time PCR 方法对广西巴马地区长寿家系(BLF,n=1301)群体外周血白细胞进行相对端粒长度(LTL)的测定,并与同地区非长寿家系(BNLF,n=283)及外地区(广西平果地区)长寿家系(PLF,n=308)、非长寿家系(PNLF,n=482)进行比较。结果(1)在整体人群及各家系人群中,LTL均与年龄呈负相关;(2)在BLF中,LTL在各年龄段均无性别差异(P>0.05),其他对照家系除个别年龄段外,也呈现类似的趋势;(3)BLF 90~年龄段的LTL长于PLF(P=0.000);(4)BLF 65~年龄段(近似于长寿老人的第一胎子女)的LTL长于BNLF(P=0.000)、PLF(P=0.011)及PNLF(P=0.021)三个对照家系,BLF 90~高龄段的LTL与三个对照家系65~年龄段的水平相当。结论长寿家系LTL明显长于普通家系,存在一定的遗传倾向。
