目的:探讨类叶升麻苷(AS)是否在体内诱导血红素加氧酶-1(HO-1)的表达。方法 AS 12.5 mg/kg 和25 mg/kg以腹腔注射的方式给予SD大鼠,在特定时间处死大鼠,取大脑皮层和纹状体组织以及肾脏和肝脏组织。提取组织蛋白,Western印迹方法检测HO-1。结果与正常对照组相比,AS可在皮层和纹状体中剂量依赖性诱导HO-1的表达上调,在外周肾脏和肝脏组织中AS剂量依赖诱导 HO-1的表达上调。结论 AS可在体内诱导HO-1的表达。%Objective To investigate whether Acteoide(AS) induce Heme oxygenase-1(HO-1) expression in vivo.Methods Male Sprague-Dawley rats(220~250 g) were treated with a single injection with 12.5,25 mg/kg body weight AS .Animals brain tissue were re-moved,and immunohistochemistry was carried out.Fresh brain,kidney and liver tissue were homogenized in lysis buffer,then the HO-1 levels were determined by Western blot.Results AS induced the expression of HO-1 in a concentration-dependent in rat cortex and striatum,also in liver and kidney.Conclusions AS could induce Heme HO-1 expression in vivo.
