丝胶对2型糖尿病大鼠肝脏胰岛素受体底物－2表达的影响

摘要

Objective To study the effects of sericin on insulin receptor substrate -2 (IRS-2) in liver of type 2 diabetes mellitus rats.Methods 36 male SD rats were randomly divided into 3 groups (n =12): normal control,diabetes model and sericin treatment groups.The type 2 diabetes model rats were induced by continuous intraperitoneal injection of streptozotocin .After successfully establishing the diabetes animal models,the rats in sericin treatment group were lavaged with sericin (2.4 g・ kg-1・ d-1 ).SP immunohistochemical stai-ning,Western blotting and RT-PCR were used to detect the IRS-2 expression in liver.Results Sericin could significantly increase IRS -2 pos-itive cell number,IRS-2 protein and mRNA expression in liver of diabetes model rats (P<0.01).Conclusions Sericin can affect insulin signaling and improve insulin resistance by up regulating IRS -2 expression in liver,thus reduce blood glucose.%目的研究丝胶对2型糖尿病大鼠肝脏胰岛素受体底物（IRS）－2表达的影响。方法36只雄性 SD 大鼠随机分为正常对照组、糖尿病模型组、丝胶治疗组，每组12只。采用链脲佐菌素连续腹腔注射法制作2型糖尿病大鼠模型。待模型成功建立后，丝胶治疗组大鼠给予丝胶（2．4 g・ kg－1・ d－1）灌胃。 SP 免疫组织化学染色、免疫印迹法和 RT－PCR 检测大鼠肝脏组织 IRS－2的表达。结果丝胶可明显提高糖尿病模型大鼠肝脏组织 IRS－2阳性细胞数、IRS－2蛋白和 mRNA 的表达（P＜0．01）。结论丝胶可通过上调糖尿病大鼠肝脏 IRS－2的表达，影响胰岛素的信号转导和改善胰岛素抵抗，起到降低血糖的作用。
