Objective To observe the effect of ethanol extract of camellia seed oil on learning and memory,and the expression of p-tau protein in brain tissue of ovariectomized AD rats.Methods 40 healthy 5 months female rats were divided into control, AD model, ca-mellia seed +AD model,estradiol+AD model groups. Camellia seed+AD model group was given camellia seed alcohol extracts of water sol-uble lavage,10 ml·kg-1· d-1(quite crude drugs 5 g/g); Estradiol + AD model group was given subcutaneous injections of estradiol 200 μg/kg,2 times per week. After 8 weeks, water maze experiment was tested. Immunohistochemical staining was used to observe the brain tissue,the brain homogenate was extracted to measure the expression of p-tau protein.Results The ability of learning and memory in AD model group was decreased obviously(P<0.01),p-tau protein deposition was decreased significantly,the amount of p-tau protein posi-tive cells was decreased significantly,the optical density value of p-tau electrophoresis was decreased significantly(P<0.01). The learning and memory ability of camellia seed+AD and estradiol+AD groups were increased significantly(P<0.01),the amount of p-tau protein pos-itive cells was increased significantly,the optical density value of p-tau electrophoresis was increased significantly (P<0.01).Conclusions Camellia seed alcohol extract has estrogen-like effect,could reduce p-tau protein,improve the learning and memory ability.%目的 探讨山茶籽醇提物对去卵巢阿尔茨海默病(AD)大鼠学习记忆、脑组织p-tau蛋白的影响.方法 SD大鼠40只随机分为假手术组、模型组、山茶籽组、雌二醇(E2)组.山茶籽组给予山茶籽醇提取物灌胃,10 ml·kg-1·d-1;E2组经皮下注射E2200 μg/kg,2次/w.假手术组和模型组给予等体积0.9%盐水灌胃(10 ml·kg-1·d-1).各组第8周大鼠称重后进行水迷宫实验;免疫组织化学染色及Western印迹法测定脑组织p-tau蛋白.结果 与假手术组比较,造模后模型组到达平台错误率显著升高(P<0.05),p-tau蛋白阳性细胞数及光密度值显著增大(均P<0.01);与模型组比较,山茶籽组及E2组错误率显著降低(均P<0.01),p-tau蛋白阳性细胞数及光密度值显著减小(均P<0.01).结论 山茶籽醇提物有雌激素样作用,可以减少去卵巢AD 模型大鼠脑组织p-tau蛋白,提高其学习记忆能力.
