有氧运动对大鼠急性心肌梗死后心脏功能的改善性研究

     

摘要

目的 观察有氧运动对大鼠急性心肌梗死后心脏功能的改善性研究.方法 实验大鼠24只,随机分为假手术组,梗死模型组、有氧运动组,每组8只.各组大鼠10周后进行血流动力学指标检测,左心室压力最大上升速率(+dp/dtmax)和左心室压力最大下降速率(-dp/dtmax),左心室收缩压(LVSP)、左心室舒张末压(LVEDP).血管紧张素Ⅱ(AngⅡ)、白细胞介素6(IL 6)、基质金属蛋白酶(MMP)-2、MMP 9、Ⅰ型胶原蛋白、Ⅲ型胶原蛋白mRNA表达.结果 与假手术组比较,梗死模型组和有氧运动组大鼠LVEDP、AngⅡ、IL-6明显升高,LVSP、+dp/dtmax、-dp/dtmax明显降低(P<0.05,P<0.01).与梗死模型组比较,有氧运动组大鼠+dp/dtmax、-dp/dtmax明显升高,LVEDP、AngⅡ、IL-6明显降低[(20.5±3.5)mm Hg vs (25.8±4.0)mm Hg,P<0.05,1 mm Hg=0.133 kPa;(1176.7±140.9)ng/L vs (1436.7±230.2) ng/L,P<0.05;(123.0±28.7)ng/L vs (161.8±14.3) ng/L,P<0.01].与假手术组比较,梗死模型组MMP-2、MMP-9、Ⅰ型胶原蛋白、Ⅲ型胶原蛋白mRNA表达明显增加(P<0.01).与梗死模型组比较,有氧运动组MMP-2、MMP-9、Ⅰ型胶原蛋白、Ⅲ型胶原蛋白mRNA表达明显降低(P<0.01).结论 有氧运动可以通过抑制血中AngⅡ、IL-6水平的增加以及抑制MMP-2、MMP-9、Ⅰ型胶原蛋白、Ⅲ型胶原蛋白mRNA表达改善心肌梗死大鼠的心脏功能.%Objective To study the effect of aerobic exercise on heart function of rats following acute myocardial infarction (AMI).Methods Twenty-four AMI SD rats were randomly divided into sham operation group,AMI model group and aerobic exercise group (8 in each group).After ten weeks,hemodynamic parameters,left ventricular+ dp/dt left ventricular-dp/dt LVSP,LVEDP,and expression levels of Ang Ⅱ,IL-6,MMP-2,MMP-9,collagen 1 and collagen 3 mRNA were measured.Results The LVEDP and serum levels of Ang Ⅱ and IL-6 were significantly higher while the LVSP,+dp/dt and-dp/dt were significanty lower in AMI model group than in sham operation group (P<0.05,<0.01).The +dp/dtmax and-dp/dtmax were significanty higher while the LVEDP and serum levels of Ang Ⅱ and IL-6 were significanty lower in aerobic exercise group than in AMI model group.The expression levels of MMP 2,MMP-9,collagen 1 and collagen 3 mRNA were significantly higher in AMI model group than in sham operation group and were significantly lower in aerobic exercise group than in AMI model group (P<0.01).Conclusion Aerobic exercise improves the heart function of rats following AMI by downregulating the expression levels of Ang Ⅱ,IL-6,MMP-2,MMP-9,collagen 1 and collagen 3 mRNA.
