Caspase信号转导通路在高糖引发牙周膜细胞凋亡中的作用

         

摘要

目的:探讨Caspase信号转导通路在高糖引发牙周膜细胞凋亡中的作用.方法:本实验采用含有不同葡萄糖浓度的培养基培养人牙周膜细胞,作用24小时后,立即使用流式细胞仪对各组凋亡细胞进行计数,并对各组细胞Caspase-3蛋白含量的变化进行检测,采用Caspase-3特异性抑制剂z-DEVD-fmR为对照.结果:高糖可以导致牙周膜细胞凋亡,并呈浓度依赖性,Caspase-3特异性抑制剂z-DEVD-fmk能够部分抑制高糖引起的牙周膜细胞凋亡.结论:高糖引发牙周膜细胞凋亡是由Caspase信号转导通路介导的.%Aim: To investigate the effect of Caspase-3 signal transduction pathway in human periodontal ligament apoptosis caused by high glucose. Methods: Human periodontal ligament cells were cultured in medium with different concentration of glucose for 24h, and then we used flow cytometer to count the percentage of apoptotic cells of each team immediately. We also detected the changes of Caspase-3 protein to determine the effect of Caspase-3 in this apoptotic process. Caspase-3 specific inhibitor z-DEVD-fmk was also used as control. Then we performed statistical analysis.Results: High glucose could induce cell apoptosis in a magnitudes dependent way. Caspase-3 specific inhibitor z-DEVD-fmk could down-regulate such effect. Conclusion: Human periodontal ligament apoptosis caused by high glucose was conducted by Caspaso-3 signal transduction pathway.
