大蒜素对Wistar鼠诱癌模型中异常腺窝病灶影响的实验研究

         

摘要

目的 通过阶段性观察大蒜素处理后Wistar大鼠诱癌模型中异常腺窝病灶(ACF)的发展情况,比较和评价大蒜素对肠道病变的预防作用.方法 80只Wistar大鼠给予二甲肼(DMH)皮下注射(20 mg/kg,每周1次,连续18周),分为大蒜素处理组和非处理组,每组分两批,分别于第8、16周处死,将美蓝染色后的大肠组织在实体镜下观察.结果 ACF均分布于中段及远端大肠(大部分位距盲肠50% ~80%处),共发现肿瘤2个,均为高分化腺癌.大蒜素处理组ACF数目较非处理组明显减少,差异有显著性统计学意义.结论 大蒜素可以明显减少Wistar鼠诱癌模型中异常腺窝病灶的数量,从而进一步证实其对结直肠癌的预防作用.%Objective To evaluate the development of aberrant crypt foci ( ACF) to tumor in the DMH-treated rats treated by the allicin. Methods 80 Wistar rats were given dimethyl hydrazine (DMH) subcutaneous injection (20 mg/ kg weekly for 18 weeks) , they were divided into allicin treated group and non-treated group, and these rats were sacrificed in 8, 16 weeks respectively. The methylene blue staining of colorectal tissue was observed in the platoscope. Results ACF located in the middle and distal colon ( most of the ACF located far from the cecum 50% to 80% ) , two tumors well-differentiated adenocarcinoma were identificated by histology. The significant difference of ACF development was found between allicin treated group and non-treated group. Conclusion Allicin can significantly reduce the development of aberrant crypt foci in Wistar rats, which is further certificated to play the role in the prevention of colorectal cancer.
