肝癌中转化生长因子(TGF)-β信号转导通路的研究进展

摘要

Hepatocellular carcinoma (HCC) is a common cancer worldwide and constitutes a therapeutic challenge with the mostly unfavorable outcome. The mechanism is still unclear, and knowledge focuses on genetic alteration resulting in loss of tumor suppressor functions, aberrant amplification and mutatinon of cancer genes. Recent study showed transforming growth factor β ( TGF-β) played an important role in HCC carcinogenesis; tumor suppressor functions at early stage of liver damage and regeneration, whereas it turned to a tumor promoter during cancer progression that exacerbates invasive and metastatic behavior. Shedding light on the mechanism will favor HCC patients with screening, diagnosis and targeting therapy at an early stage.%肝细胞肝癌( HCC)是常见的恶性肿瘤,严重威胁人类健康.其发病机制仍不清楚,已有的研究集中在遗传学改变导致的肿瘤抑制基因功能缺失、癌基因突变和异常扩增方面,进一步研究则表明转化生长因子β( transforming growth factor β,TGF-β)信号转导通路在其中发挥着双重调控作用:在肿瘤发生的起始阶段TGF-β扮演着肿瘤抑制者的角色,而在进展期TGF-β起着促使肿瘤细胞浸润转移的作用.对其作用机制的深入阐述对肝癌的早期筛查、早期诊断、靶向治疗有很重要的意义.
