姜黄素联合顺铂对宫颈癌Hela细胞化疗敏感性的影响及机制探讨

摘要

Objective To investigate the influence and mechanism of curcumin combined cisplatin on the chemosensitivity of Hela cells, and to provide reference for clinical practices. Method The cervical cancer Hela cells were selected for In vitro development, curcumins with different concentrations (compatible with cisplatin) were utilized to intervene cervical cancer Hela cells, MTT method was used to determine the proliferation inhibition rate of medicine on Hela cells, RT-PCR and Western blot method were utilized to determine expression details of p53 protein, the influence and mechanism of curcumin combined cisplatin on cervical cancer Hela cells chemotherapy sensitivity were explored. Result The inhibition rate and rate of apoptosis-inducing against the proliferation of cervical cancer Hela cells in the experimental group were signiifcantly higher than control group and curcumin group (P<0.05). Expressions of COX-2, Caspase-3 and Maspin in experimental group were signiifcantly better than control group and curcumin group (P < 0.05). Expressions of p53 proteins within 24 hours upon the action of curcumin with different concentrations increased with the increase in the concentration of with curcumin, and the difference was statistically signiifcant (P<0.05). Conclusion The curcumin combined cisplatin delivers signiifcant inlfuences on the chemosensitivity of Hela cells, it can inhibit the proliferation of Hela cells and induce the apoptosis of cellsand increase the sensitivity, which may be realized by increasing expressions of p53 gene.%目的:研究姜黄素联合顺铂对宫颈癌Hela细胞化疗敏感性的影响以及机制,为临床实践提供参考和借鉴。方法选用人宫颈癌Hela细胞进行体外培养,利用不同浓度姜黄素(配伍顺铂方案)干预人宫颈癌Hela细胞,采用MTT法检测药物对宫颈癌Hela细胞的增殖抑制率;采用RT-PCR和Western blot法检测p53蛋白的表达情况,探讨姜黄素联合顺铂方案对宫颈癌Hela细胞化疗敏感性的影响以及机制。结果实验组对宫颈癌Hela细胞增殖的抑制率、诱导凋亡率明显高于对照组及姜黄素组(P<0.05);实验组COX-2、Caspase-3、Maspin的表达情况明显优于对照组及姜黄素组(P<0.05);不同浓度姜黄素作用24小时后,p53蛋白的表达随姜黄素浓度的增加而增加,差异具有显著性(P<0.05)。结论姜黄素联合顺铂对宫颈癌Hela细胞化疗敏感性具有重要影响,可以抑制宫颈癌Hela细胞的增殖并诱导细胞凋亡,能够增加其敏感性,这可能是通过增加p53基因的表达而实现的。
