Objective To develop a method for the determination of amitriptyline in rat blood and urine by gas chromatography (GC).Methods The amitriptyline standard and internal standard was added to blank samples of blood and urine.The experimental rats were given amitriptyline in double dosage of the LD50 with intragastric administration.Blood and urine were extracted from poisoned rats.Amitriptyline was extracted from samples with diethyl ether and for quantitative analysis by GC/FID.Experimental conditions were studied.Results To analyze amitriptyline in blood and urine with this method, the linear range was 5 ~ 150μg/mL (r = 0.993 ) and 5 ~ 150μg/mL ( r = 0.998 ) respectively.The limit of detection was 1 μg/mL and 1μg/mL (S/N≥3).The RSD were less than 6%.The recovery rate was about 95.6% ~ 105.6%.Conclusion This method has shown simple and accurate for determination of amitriptyline.%目的 建立血液及尿液中阿米替林(AMTL)的气相色谱分析方法.方法 以正常大鼠血液及尿液为空白样本,分别添加AMTL标准品和内标SKF(525A).实验大鼠以AMTL 2倍LD(50)灌胃,致大鼠急性中毒后提取血液及尿液.用乙醚提取样本中AMTL,采用GC/FID法进行定量分析,并考察实验条件.结果 采用该方法,血液及尿液中AMTL线性范围分别为5~150μmL(r=0.993)和5~150μg/mL(r=0.998);最低检测限(S/N≥3)均为1.0μmL;日内、日间精密度均小于6%,回收率在95.5%~105.6%之间.结论 该方法方操作便捷、准确度高,适用于AMTL临床治疗中血药浓度快速监测和法医毒物分析鉴定.
