神经细胞粘附分子对恶性胶质瘤细胞体外侵袭能力的影响

摘要

Objective To investigate the effect of a transmembrane isoform of neural cell adhesion molecule (NCAM) on the invasive abilitity of malignant glioma cells in vitro. Methods Plasmid containing NCAM cDNA fragment was transfected into the NCAM-negative U251MG cells by lipofectamine. After transfection,the selected clones of NCAM positive cells were cultured in Boyden chamber for 7 h. The invasion of cells was assessed by calculating the number of cells penetrating through a matrigel-coated filter. And the monoclonal anti-NCAM antibody(1∶30 diluted)was added into the culture medium of NCAM-positive glioma cells to study its effect on the invasion of glioma cells. Result The invasion of NCAM-negative U251MG was stronger (223.67±40.28) than that of NCAM-positive U87MG cell strain (70.00±20.95)(P<0.01).Transfection with NCAM cDNA (103.00±37.00) reduced the invasion of U251MG (223.67±40.28)(P<0.05). After the treatment using monoclonal antibody (anti-NCAM),the invasion of U87MG cells and transfected U251MG cells were enhanced (P<0.05).Conclusion NCAM could inhibit the invasion of malignant glioma cells in vitro.%目的 探讨神经细胞粘附分子(NCAM)对恶性胶质瘤细胞侵袭行为的影响。方法 用脂质体法将NCAM cDNA转染人胶质瘤细胞株U251MG,体外波艾登(Boyden)小室中培养7 h,同时用抗NCAM单克隆抗体(1∶30稀释)封闭细胞表面的NCAM作对照,观察转染前后U251MG细胞及U87MG细胞侵袭基质凝胶能力的变化规律。结果 转染后U251MG细胞表达NCAM,侵袭能力(103.00±37.00)较转染前(223.67±40.28)减弱(P<0.05),对照组U87MG细胞体外侵袭能力(70.00±20.95)亦比转染前U251MG细胞低(P<0.01),经抗NCAM单克隆抗体封闭细胞表面NCAM抗原后,U87MG细胞及转染后U251MG细胞侵袭力均比抗体处理前增强(P<0.05)。结论 NCAM对恶性胶质瘤细胞体外侵袭能力起抑制作用。
