类固醇受体共激活因子-3在人肝癌组织中的表达及其临床意义

摘要

Objective To investigate the expression of steroid receptor coactivator-3 (SRC-3) in human hepatocellular carcinoma and its relation with the clinicopathologic characteristics of the carcinoma.Methods Immunohistochemistry and reverse transcriptase-polymerase chain reaction (RT-PCR) were used to detect the level of protein expression and mRNA tanscription of SRC-3 in hepatocellular carcinoma tissues and the paired paratumorous liver (PTL) tissues from 94 patients with hepatocellular carcinoma.By combining with clinical features of these patients,the relationship between SRC-3 expression and clinieopathologic characteristics was studied statistically.Results Immunohistochemistry showed that the positive expression rate of SRC-3 protein was higher in hepatocellular carcinoma tissues than in their paired PTL tissues (55.3％ vs.10.6％,P ＜0.05).SRC-3 mRNA transcription level in hepatocellular carcinoma tissues was significantly higher than that in the PTL tissues (P ＜ 0.05).The clinicopathologic evaluation revealed that the higher protein expression and mRNA transcription level of SRC-3 in hepatocellular carcinoma tissues was correlated with hepatitis B virus (HBV) infection,serum α-fetoprotein levels and pathologic grade (P ＜ 0.05).Conclusion SRC-3 is overexpressed in hepatocellular carcinoma and closely correlated with the clinicopathologic features of the carcinoma.%目的 探讨类固醇受体共激活因子-3(SRC-3)在人肝癌组织的表达及其与肝癌临床病理特征之间的关系.方法 采用免疫组织化学法和逆转录-聚合酶链反应(RT-PCR)法分别检测SRC-3基因在94例肝癌组织及其癌旁组织的蛋白表达水平和mRNA转录水平,分析SRC-3的表达与临床病理特征的相关性.结果 SRC-3蛋白在肝癌组织中阳性表达率为55.3％ (52/94)；相对应的癌旁肝组织则较少见SRC-3蛋白表达,其阳性率为10.6％ (10/94),且阳性染色分布散在、稀疏.肝癌组织中SRC-3 mRNA的转录水平亦高于其相对应的癌旁肝组织(P＜0.05).SRC-3蛋白表达水平和SRC-3 mRNA转录水平均与肝癌患者乙型肝炎病毒(HBV)感染、甲胎蛋白(AFP)水平、病理分级等因素明显相关(P＜0.05).结论 SRC-3基因在人肝癌组织的表达水平显著高于癌旁组织,SRC-3基因高表达与肝癌的临床病理特征密切相关.
