微小RNA-34a在结直肠癌患者血清及组织中的表达及其与E盒结合锌指蛋白2表达的相关性

摘要

Objective To investigate the expressions of microRNA (miRNA,miR)-34a in serum and tissue of patients with colorectal cancer and its correlation with the expression of zinc finger E-box binding protein 2 (ZEB2).Methods Fifty patients with colorectal cancer undergoing surgical treatment in Henan Tumor Hospital were selected from January 2012 to June 2012.Meanwhile,100 healthy people were selected as control group.The expression of miR-34a in serum of colorectal cancer patients and the control group,and that of miR-34a and ZEB2 in tumor tissues were detected by real-time reverse transcriptase-polymerase chain reaction(RT-qPCR).The expression of ZEB2 protein in tumor tissues was tested by Western blotting.Results The relative expression level of miR-34a in the serum of patients with colon cancer was (0.94 ±0.13),significantly lower than (1.48 ±0.52) of the control group (P ＜ 0.05).The relative expression level of miR-34a in colorectal cancer tissues was statistically significantly lower than that in the adjacent tissues and normal colorectal tissues (P ＜ 0.05).The expression of miR-34a in the serum of colorectal cancer and colorectal cancer tissues was correlated with lymph node metastasis and distant metastasis (P ＜ 0.05).The relative expression levels of ZEB2 mRNA and protein in colorectal cancer tissues were higher than the adjacent tissues and normal colorectal tissues (P ＜ 0.05).Pearson correlation analysis showed that the expression of miR-34a in colorectal cancer tissue was negatively correlated with the expression of ZEB2 protein (r =-0.301,P ＜ 0.05).Conclusion The expressions of miR-34a in serum and tissue of patients with colorectal cancer were low.It might be negative regulation ZEB2 to achieved effectthe metastasis and invasion of tumor cell.%目的 探讨微小RNA(miRNA,miR)-34a在结直肠癌患者血清及肿瘤组织中的表达及其与E盒结合锌指蛋白2(ZEB2)表达的相关性.方法 选取住院行手术治疗的结直肠癌患者50例,同时选取100例健康体检者作为对照组,利用实时定量反转录聚合酶链反应(RT-qPCR)检测结直肠癌患者和对照组血清中miR-34a表达,并检测肿瘤组织中miR-34a及ZEB2表达,利用Westernblot检测ZEB2蛋白在肿瘤组织中的表达.结果 miR-34a在结直肠癌患者血清中相对表达量为0.94±0.13,远低于对照组血清中相对表达量(1.48±0.52),差异有统计学意义(P＜0.05);miR-34a在结直肠癌组织中的相对表达量低于癌旁组织和正常结直肠组织,差异均有统计学意义(P＜0.05);miR-34a在结直肠癌患者血清和结直肠癌组织中的表达量均与淋巴结转移和远处转移有关(P＜0.05);ZEB2 mRNA和蛋白在结直肠癌组织中相对表达量均高于癌旁组织和正常结直肠组织,差异均有统计学意义(P＜0.05);Pearson相关分析显示,结直肠癌患者组织中miR-34a表达量与ZEB2蛋白表达量呈负相关(r=-0.301,P＜0.05).结论 miR-34a在结直肠癌患者血清及肿瘤组织中均呈低表达,可能通过负向调控ZEB2而实现对肿瘤细胞转移和侵袭的影响.
