慢性束缚心理应激对大鼠血管内皮结构和功能的影响

             

摘要

目的 探讨慢性束缚心理应激对大鼠血管内皮结构和功能的影响及其机制.方法 20只清洁级Wistar大鼠随机分为对照组与模型组,采用重复束缚法6周建立心理应激反应模型,放免法检测血浆内皮素(ET),硝酸还原酶法检测血清NO;循环酶法测定血清同型半胱氨酸(Hcy)水平;光镜观察胸主动脉形态学变化,透射电镜观察主动脉组织内皮细胞超微结构变化;免疫组化法检测主动脉组织中内皮素-1(ET-1)的表达.结果 模型组大鼠血浆ET含量及血清Hcy含量较对照组明显升高,血清NO明显下降(P<0.05或P<0.01);主动脉组织中ET-1高度表达;内皮细胞超微结构明显改变.结论 慢性束缚心理应激可引起大鼠明显的血管内皮结构和功能的损伤,其机制可能与血清Hcy水平升高有关.%Objective To investigate the influence on the construction and function in vascular endothelial of rats with chronic constraint psychological stress and explore mechanism. Methods Twenty healthy male Wistar rats of sanitary degree were randomly divided into the normal control group and the psychological stress model group. To establish the chronic psychological stress model by repeated restraint for 6 weeks. The content of plasma endothelin ( ET ) was detected with radio-immunoassay, serum nitro oxygen ( NO ) was detected with nitrate reductase, and serum Hey with enzymatic cycling assay. The morphological changes of thoracic aorta was observed with light microscope and the ultramicrostructure changes of aorta was observed with transmission electron microscope. Immunohistochemistry method was applied to analyze the expression of endothelin-l( ET-1 ) in aorta tissue. Results The content of ET in plasma and Hey in serum of model rats increased signifi-cantly( P < 0.05 ). The content of NO in serum of model rats lowered than that in control group significantly( P < 0. 01 ). The expression of ET-1 in the arterial tissue of model rats enhanced. The ultramicrostructure of vascular endothelium changed obviously. Conclusion The chronic constraint psychological stress could cause endothelial dysfunction, the mechanism may be up-regulating of serum Hey.
