rhHSP70对食管癌细胞RNA转染的脐血树突状细胞诱导CTL抗肿瘤功能的影响

         

摘要

Objective To investigate the effect of rhHSP70 and human cord blood dendritic cells( DCs ) transfect with RNA of esophageal carcinoma on anti-tumor activity of CTL. Methods The RNA of T. Tn cells were prepared by Trizol reagent. The cord blood monocytes were cultured to produce DCs with rhSCF, rhGM-CSF and IL-4. The DCs were collected and transected with tumor cell RNA. CpG ODN was added in culture medium to enhance the DCs vaccine. The mixed lymphocyte reaction ( MLR ) and cytotoxicity of CTL to T. Tn cells was assayed by MTT colorimetry. Results Compared with pretransfect stage, DCs transfect with RNA of esophageal carcinoma cells showed more MHC-Ⅰ, MHC-Ⅱ, CD54, CD 80 and CD86 ( P < 0.01 ). Compared with immature DCs, mature DCs transfect with tumor RNA enhanced tumor cytolytic activities of T lymphocyte. rhHSP70 could significantly increase the number of molecules on surface of activated T lymphocyte. Conclusion The vaccine of cord blood DCs with adjuvant of rhHSP70 can provide effective and specific way for immunological therapy of esophageal carcinoma.%目的 研究重组人热休克蛋白70(rhHSP70) 联合食管癌细胞RNA转染脐血树突状细胞(DCs)对细胞毒性T淋巴细胞(CTL)特异性抗肿瘤作用的影响.方法 分离脐血单个核细胞,经rhSCF、rhGM-CSF和IL-4诱导和食管癌RNA转染形成成熟DCs,加入rhHSP70,检测DCs表面标志及DCs诱导的T细胞增殖、CTL杀伤活性.结果 食管癌RNA转染后DCs高表达抗原提呈分子、协同刺激分子和黏附分子,对T细胞的促增殖作用和CTL杀伤活性显著增强(P<0.01),rhHSP70能显著促进DCs功能.结论 rhHSP70具有增强食管癌细胞RNA转染的DCs对T细胞的促增殖和CTL杀伤活性的作用.
