参蛇洗剂对小鼠变应性接触性皮炎模型的药效学及机制研究

             

摘要

Objective To investigate the effects of Shenshe lotion on mice models of allergic contact dermatitis (ACD) and to explore its possible therapeutic mechanism. Methods 60 BALB/c mice were randomly divided into six groups: blank control group, model control group, Shenshe lotion of high to low concentrations (1:2, 1:1 and l:0.5) groups and positive control group (1% hydrocortisone solution). Ear thickness and allergic reaction score were measured before stimulation and 12, 48 h after stimulation respectively, and scratching frequency within one minute was collected. Meanwhile, levels of inflammatory cytokines such as tumor necrosis factor-α (TNF-α), interferon-γ (IFN- γ), interleukin-1β (IL-1β) and interleukin-4 (IL-4) in peripheral blood were detected, and pathological changes such as inflammatory cell infiltration in ear skin were observed by hematoxylin and eosin (HE) staining. Results Shenshe lotion of different concentrations could alleviate the ear thickness, allergic reaction score and scratching phenomenon in ACD mice, and it could reduce the inflammatory degree of their ear skin. There was no statistical difference among different concentrations of Shenshe lotion group and 1% hydrocortisone lotion group in overall therapeutic effect. Meanwhile, Shenshe lotion alleviated the scratching frequency markedly than 1% hydrocortisone did. The levels of TNF-α and IFN-γ decreased significantly in peripheral blood of ACD mice (P<0.01), while IL-1β and IL-4 levels increased (P<0.01). And these cytokine levels didn't change significantly after the treatment of Shenshe lotion of different concentrations or 1% hydrocortisone lotion. Conclusion By having a similar overall therapeutic effect and a better anti-itching effect than 1 % hydrocortisone solution, Shenshe lotion might have a good therapeutic effect on ACD mice. Further researches on the effects of Shenshe lotion on inflammatory cytokines of ACD mice are required, both in peripheral blood and skins.%目的 观察参蛇洗剂对小鼠变应性接触性皮炎(allergic contact dermatitis,ACD)模型的药效学并初步探究其可能治疗机制.方法 60只BALB/c小鼠随机平分为6组:空白对照组、模型对照组、高中低3个剂量参蛇洗剂组(1:2,1∶1和1∶0.5)及阳性药对照组(1%氢化可的松溶液).在激发前、激发后12h和激发后48 h分别对小鼠耳厚度及耳变态反应程度进行测量评分,计数1 min内小鼠搔抓耳部次数,采集外周血检测炎症因子肿瘤坏死因子α(tumor necrosisfactor-α,TNF-α)、γ干扰素(interferon-γ,IFN-γ)、白细胞介素1β(interleukin-1β,IL-1β)及白细胞介素4(interleukin--4,IL-4)水平,并以苏木精-伊红(hematoxylin and eosin,HE)染色观察小鼠耳部皮肤炎性细胞浸润等病理变化情况.结果 不同浓度的参蛇洗剂均能减轻小鼠ACD模型的耳厚度和变态反应评分,缓解小鼠搔抓现象,减轻耳部皮肤炎症程度,总体治疗上各浓度组及1%氢化可的松溶液组差异无统计学意义,而小鼠搔抓耳部次数明显少于1%氢化可的松溶液组.ACD小鼠外周血中TNF-α和IFN-γ水平明显下降(P<0.01),IL-1β和IL-4水平明显增高(P<0.01).给予不同剂量参蛇洗剂及1%氢化可的松溶液外涂后,其外周血上述中细胞因子水平变化均不明显.结论 参蛇洗剂对ACD可能具有较好的治疗作用,在总体治疗作用上和1%氢化可的松溶液相当,在抗瘙痒上明显优于1%氢化可的松溶液.参蛇洗剂对ACD小鼠外周血及皮肤局部炎症因子的影响仍待进一步研究.
