白细胞介素-10对体外培养人牙囊细胞MCP-1表达的影响

AIM:To study the effect of IL-10 on the expression of monocyte chemotactic protein-1(MCP-1)in human dental follicle cells(HDFCs).METHODS:HDFCs of the 5th passage were CO-cultured with 25ng/mL IL-10 for 0,1,3,6,9 and 12 h.The mRNA expression of MCP-1 Waft detected by RT-PCR.In the next experiment the cells were CO-cultured with 25ng/mL IL-10 for 0,2,4,6 and 8h.The protein expression of MCP-1 wag examined by ELISA.RESULTS:25ng/mL IL-10 down-regulated the mRNA expression of MCP-1 in 1-6h(P<0.05)and the protein expression of MCP-1 in 4-8h(P<0.05).CONCLUSION:IL-10 inhibits MCP-1 expression in HDFCs.Since MCP-1 is an important factor to recruit mononuclear cells.IL-10 suppresses the recruitment of these cells to the follicle by down-regulating MCP-1 expression.%目的:研究白细胞介素-10(Interleukin-10,IL-10)对体外培养人牙囊细胞单核细胞趋化蛋白-1(Monocyte chemotactic protein-1,MCP-1)表达的影响.方法:体外培养人牙囊细胞,取第5代细胞加入25 ng/mL IL-10分别作用0、1、3、6、9、12 h,用RT-PCR法检测MCP-1mRNA表达的变化.取第5代人牙囊细胞加入25 ng/mL IL-10分别作用0、2、4、6、8 h,用ELISA法检测上清液中MCP-1蛋白的含量.结果:MCP-1mRNA表达在1-6h降低(P<0.05),蛋白分泌在4-8 h减少(P<0.05).结论:IL-10抑制体外培养人牙囊细胞中MCP-1的表达.MCP-1是重要的单核细胞趋化因子,因此IL-10通过降低MCP-1的表达对单核细胞进入牙囊发挥抑制作用.
