五倍子水提取物抑制粪肠球菌粘附能力的体外实验

         

摘要

目的:观察五倍子水提取物预处理对于粪肠球菌粘附能力的影响.方法:①取3块底部铺被Ⅰ型胶原蛋白的细胞培养板,1块经200 mg/mL五倍子预处理后,接种粪肠球菌;其余两块不用五倍子预处理,1块直接接种粪肠球菌,另1块仅用PBS冲洗用于空白对照.置于37 ℃厌氧条件下培养2 h后,进行结晶紫染色,并用酶联仪检测各孔600 nm波长处的吸光度值;②取消毒后的牛牙中段根管样本8个,将其随机分为实验组(200 mg/mL五倍子溶液处理)和对照组(生理盐水处理)(n=4);两组一并置于37 ℃水浴锅中处理2 d后,再浸入粪肠球菌悬液中37 ℃厌氧条件下连续孵育28 d;用扫描电镜分别观察各样本根管壁上的细菌生长情况.结果: ①酶联仪检测显示,五倍子预处理组的吸光光度值明显低于未处理组(P<0.05),3组间两两相比均有统计学差异(P<0.05);②扫描电镜观察结果显示,经五倍子预处理组的牙本质表面所粘附的细菌量明显少于未处理组.结论:五倍子预处理胶原蛋白和牙本质可明显降低粪肠球菌对二者的粘附能力.%AIM:To assess the effect of aqueous extract of Galla chinensis on the adhesion ability of E.faecalis in vitro.METHODS:The effect of Galla chinensis on the adhesion ability of E.faecalis to SigmaScreen collagen I coated plates was evaluated by pretreatment of the coating for 1 h before E.faecalis were seeded on and by the comparison between experimental and control groups.Then the effect of Galla chinensis on the adhesion ability of E.faecalis to dentin was observed by scanning electron microscope using an in vitro root canal infection model.RESULTS:Galla chinensis extracts significantly decreased the adhesion of E.faecalis to collagen I coated plates and bovine dentinal tubules (P<0.05).CONCLUSION::Galla chinensis can inhibite the adherence of E.faecalis to collagen or dentin.
