电压依赖性钾通道和钙激活钾通道在高血压大鼠外周血淋巴细胞的变化

         

摘要

Objective To determine express of Kvl. 3,KCa3. 1 mRNA in lymphocyte derived from sponsta-neously hypertensive rat (SHR) and identify the relationship between lymphocyte Kvl. 3,KCa3. Land hypertension. Methods Male SHRs and normotensive Wistar rats were obtained from Experimental Animal Company of Beijing Vital River and Experimental Animal Center of Xinjiang Medical University. Peripheral blood were collected from SHRs and normotensive Wistar rats. Peripheral lymphocytes were separated from heparinized whole blood by standard Ficoll-Hypaque density gradient centrifugation. Real-time PCR was used to detect the expression of Kvl. 3,KCa3. 1 mRNA in lymphocytes. Results ( 1) The expression levels of Kv 1.3 mRNA in lymphocytes from SHRs were significantly increased compared with Wistar rats (P<0.05). (2)Compared with the controls, Kca 3. 1 mRNA were incresed in in lymphocytes from SHRs (P<0.05). (3)Linear correlation existed between systolic blood pressure and expression of Kvl. 3,Kca3.1 mRNA in lymphocytes from SHRs ( r - 0. 830,P = 0.003;r = 0.674,P = 0.03,respectively). Conclusion The lymphocyte Kvl. 3 ,KCa3. 1 mRNA expression are upregulated in SHR,which suggests that Kvl. 3 and Kca 3. 1 maybe contribute to the development of hypertension.%目的 研究自发性高血压大鼠(SHR)外周血淋巴细胞电压依赖性钾通道(Kv)、钙激活钾通道( KCa) mRNA表达的变化,探讨淋巴细胞Kv1.3、KCa3.1在高血压病中的变化.方法 使用密度离心法采集10只SHR和10只正常Wistar大鼠外周血淋巴细胞,运用实时荧光定量PCR技术检测外周血淋巴细胞Kv1.3、KCa3.1 mRNA表达水平.结果 (1)SHR组Kv1.3 mRNA表达水平高于对照组(P<0.05);(2)与对照组相比,SHR组KCa3.1 mRNA表达水平增高,差异有统计学意义(P<0.05).(3)SHR收缩压和淋巴细胞Kv1.3、KCa3.1基因表达呈正相关:相关系数为r=0.830,P=0.003;r =0.674,P=0.03.结论 SHR淋巴细胞Kv1.3、KCa3.1 mRNA表达增高,推测淋巴细胞钾通道增多可能与高血压病的发生发展相关.
