目的:探讨柚皮素对气道黏液高分泌的作用和相关分子机制.方法:培养A549细胞株,构建气道黏液高分泌模型,以HNE为刺激因素,柚皮素和DMTU为干预因素,观察细胞内ROS含量、EGFR、P-EGFR及MUC 5AC的表迭变化.结果:ROS含量的变化与HNE浓度成正比,柚皮素预处理后ROS明显减少(P<0.01);50nmoL/L的HNE组中,EGFR mRNA、MUC5AC mRNA和蛋白及P-EGFR蛋白均较对照组显著增加(P<0.05);各干预组中上述指标均下调(P<0.05),差异均有统计学意义.结论:柚皮素具有抑制气道黏液高分泌的作用.%Objective: To investigate the effects of Naringenin on airway mucous hypersecretion and the relative molecular mechanism.Methods: The cell model of mucous hypersecretion was made by culturing A549 cell lines.Results: A dose-dependent increasing of ROS production in cells exposed to HNE was detected, after the cells were pre-treated with Naringenin, the content of ROS, EGFR mRNA, MUC5AC mRNA and P-EGFR proteins were decreased significantly as compared with the 50nmol/L HNE group ( P < 0.01 ).After the cells were pre-cultured with Naringenin, DMTU and Naringenin + DMTU, respectively, The above measurements were decreased significantly as compared with the HNE group (P < 0.01 ).Conclusions: Naringenin attenuated airway mucous hypersecretion under inflammation.
