Objective: The present study was to investigate the learning memory and anti-oxidative effects of Wuzi Yanzong Fang on D-galactose-induced senescence. Methods: Eight-week-old male mice were divided into four groups of vehicle-treated group, D-galactose-treated group, Wuzi Yanzong Fang (0. 49g/kg、0. 98g/kg) groups. Mice were injected with D-galactose (150mg/kg/d) subcutaneously and intragastrically administered Wuzi Yanzong Fang simultaneously. After 8 weeks of treatment, the animal behavior and jumping stand were tested. Cerebral index, spleen index and thymus index were measured. The SOD activity and MDA content were evaluated by biochemistry detection. Results: Wuzi Yanzong Fang markedly extended the time of staying in the target quadrant and increased the number of times through the platform. The number of errors in jumping stand experiment decreased significantly compared with D-gal-treated group. Meanwhile, Wuzi Yanzong Fang alleviated the atrophy of brain and thymus in mice, elevated activity of serum superoxide dismutase, and decreased malondialdehyde content. Conclusion: Wuzi Yanzong Fang can improve learning and memory of aging mice induced by D-gal and its mechanism, at least in part, via modification of the redox system in the organism.%目的:探讨五子衍宗方对D-gal致衰老小鼠学习记忆及抗氧化损伤的影响.方法:小鼠随机分为4组,模型组及给药组皮下注射D-gal,正常组皮下注射NaCl;同时各组给予相应的药物治疗,8周后检测小鼠的学习记忆能力、小鼠的脑、脾脏及胸腺指数、血清中SOD和MDA水平.结果:五子衍宗方能延长小鼠目标象限停留时间,增加穿过平台次数,降低跳台错误次数;降低脑及胸腺重量指数,增加SOD活性,降低MDA含量.结论:五子衍宗方能改善衰老小鼠的学习记忆能力,其机制可能与提高小鼠的抗氧化能力有关.
