To study the effects and mechanisms of Gymnema sylvestre on improvement of insulin resistance in adipose tissue of KKAy mice.Methods:18 KKAy mice were divided randomly according to body weight level into Models (DM) Gymnema sylvestre group (GS) and 9 normal C57BL/6J mice as normal control group(NC).Fasting plasma glucose(FPG ) of all mice were tested at every weekend,and at the end of the experiment all animals were tested FPG,Fasting insulin level (Fins)for evaluating Insulin Sensitivity Index (ISI).The expressions of PPAR-γmRNA 、PI3 κ-p85mRNA、CAPmRNA and GluT-4mRNA were determined by Real-time PCR.Results:Compared with DM group,Gymnema sylvestre down-regulated the levels of FPG,Fins,up-regulated ISI and the expressions of PPAR-γmRNA、PI3 κ-p85mRNA、CAPmRNA and GluT-4mRNA in adipose tissue.Conclusion:Gymnema sylvestre improved insulin resistance of KKAy mice by increasing the expressions of PPAR-γmRNA、PI3κ-p85mRNA、CAPmRNA and GluT-4mRNA in adipose tissue.%目的:探讨匙羹藤(GS)改善KKAy小鼠脂肪组织胰岛素抵抗(IR)的作用机制.方法:KKAy小鼠分为模型组(DM)和GS组,C57BL/6J小鼠为正常组(NC),给药8周后测定血清胰岛素(Fins)、胰岛素敏感指数(ISI)、过氧化物酶增殖体受体-γ(PPAR-γ)、PI3 κ-p85、Cb1衔接蛋白(CAP)、葡萄糖转运蛋白4(GluT-4) mRNA水平.结果:与DM比较,FPG、Fins降低,ISI升高,PPAR-γmRNA、PI3κ-p85mRNA、CAPmRNA、GluT-4 mRNA表达量升高.结论:GS可上调脂肪组织PPAR-γ、PI3 κ-p85、CAP、GluT-4mRNA的表达量,改善IR.
