FOXO3a表达水平与大鼠睾丸间质Leydig细胞凋亡的相关性初步研究

         

摘要

目的 探讨大鼠睾丸Leydig细胞中FOXO3a (forkhead box O 3a)转录因子表达水平与Leydig细胞凋亡的相关性.方法 取出生后2d和3月龄大鼠的睾丸,用免疫组化方法观察FOXO3a和Bim在睾丸组织中的表达.结果 在出生2d的新生大鼠睾丸内,FOXO3a在大部分Leydig细胞胞核和胞浆内表达而只在少数Leydig细胞的胞核内高表达,Bim也在大部分Leydig细胞胞核和胞浆内表达.在3月龄的成熟大鼠睾丸内,FOXO3a在大部分Leydig细胞的胞核内高表达,Bim也在大部分Leydig细胞胞核和胞浆内表达.结论 出生后2d和3月龄的大鼠睾丸内,FOXO3a在部分Leydig细胞核内高表达,提示FOXO3a可能参与新生鼠与成年鼠睾丸内Leydig细胞的凋亡调控;Bim同时也在大部分Leydig细胞胞核和胞浆内表达,提示FOXO3a可能通过调节下游的Bim来调节Leydig细胞的凋亡.%Objective To study the correlation between FOXO3a expression and Leydig cell apoptosis in rat's testes. Methods Testes of rats aged 2 days and 3 months were collected . The protein expression of FOXO3a and Bim was examined by immunohistochemistry. Results For testes of rats aged 2 days t, FOXO3a was mostly detected in both the nuclei and the cytoplasm of Leydig cells and overexpression of F0X03a was detected in the nuclei of Leydig cells too, Bim was mostly detected in both the nuclei and the cytoplasm of Leydig cells. For testes of rats aged 3 months, overexpression of FOXO3a was detected in the nuclei of Leydig cells and Bim was mostly detected in both the nuclei and the cytoplasm of Leydig cells. Conclusion For testes ofrats of both age FOXO3a is highlyexpressedinthe nuclei of some Leydig cells, suggesting that FOXO3a might be involved in the regulation of Leydig cell apoptosis. Additionally, Bim is expressed in some Leydig cells, suggesting that FOXO3a might regulate Leydig cell apoptosis via Bim.
