应用PCR技术快速检测口腔念珠菌菌株

摘要

Objective This study was performed to through PCR technology to find a rapid, accurate and effective detection that is used to detect oral Candida. Methods Collecting 100 specimens of saliva, then culture these on CHROMagar media and detect by PCR. Results The positive rate of oral Candida is 35% by traditional cultural detection while by PCR technology in two primers are 68% and 49% respectively. The traditional detections for Candida are time -consuming,especially in culture method which consumes 48hrs at least;to apply PCR method to detect Candidas only need 5hrs with high positive rate and accuracy. Conclusion To compare with traditional culture method for Candidas,PCR technology is more perfect and handy.%本研究将PCR技术应用于口腔念珠菌检测中,寻找出一种能够快速、准确、有效检测口腔念珠菌的方法.方法:收集口腔临床唾液样本100例,科玛嘉显色培养基进行培养,同时应用PCR方法对100例临床样本进行检测鉴定.结果:传统念珠菌培养方法的检出率为35％；应用两对引物,PCR方法的检出率分别为68％和49％.传统的念珠菌检测方法耗时长,特别是培养法,需时48hrs；应用PCR方法对念珠菌进行检测,只需要5hrs,且阳性检出率和准确性较高.结论:PCR方法快速敏感、特异性高,相较于念珠菌传统培养方法,是一种更加简便理想的检测方法.
