胶原海绵为载体构建大鼠羊膜上皮细胞复合体的实验研究

         

摘要

Objective To investigate the growth of rat amniotic epithelial cells (AECs) in the collagen sponge when the cells implant to the scaffolds after they were cultured in vitro.Methods The 3th SD rats AECs were seeded into the collagen scaffold.To observe the growth and proliferation of cells before and after planting with Light microscopy and method of fluorescence labeling.ELISA was taken to detect the angiogenesis factors VEGF,bFGF,TGFβ in the supernatant fluid of the AECs complex.The method of Fluorescently labeled was taken to observe the survival of the cells after transplantation.Results Rat AECs in collagen sponge scaffolds grown well,the content of VEGF,bFGF,TGFβ in the supernatant fluid increased gradually.Cells still survived well after AECs/collagen sponge complex transplanting in vivo.Conclusion AECs grow well in the Collagen Sponges and transplanting the AECs/collagen sponge complex in vivo,which lay the foundation of cell therapy.%目的:大鼠羊膜上皮细胞(AECs)体外培养并种植于胶原海绵形成细胞支架复合体后,探讨细胞在支架上分别于体内及体外时的生长情况.方法:取第三代AECs细胞种植于胶原海绵支架.光镜下及用荧光标记法观察细胞种植前后的增殖情况;ELISA法检测细胞支架上清液中VEGF、bFGF、TGF β的含量.并通过荧光标记示踪法观察细胞支架植于体内后细胞的存活情况.结果:AECs在胶原海绵支架上生长情况良好,细胞支架上清液中VEGF、bFGF、TGFβ的含量随时间增长呈不断上升趋势,将细胞支架移植于体内后经过观察发现细胞仍存活良好.结论:AECs在胶原海绵支架上及体内移植后生长情况良好,这为利用AECs胶原海绵支架复合体进行细胞治疗奠定了实验基础.
