维吾尔药异常黑胆质成熟剂总黄酮对增生性瘢痕成纤维细胞增殖及胶原蛋白表达的影响

摘要

目的:探讨维吾尔药异常黑胆质成熟剂总黄酮(Abnormal savda munziq-total flavonoids,ASMq-TF)对体外培养的增生性瘢痕成纤维细胞(Hypertrophic scar fibroblasts,HFs)增殖抑制作用及对相关胶原蛋白合成的影响.方法:对HFs进行体外培养并给予不同浓度的ASMq-TF处理,然后,通过CCK-8检测其细胞增殖情况,通过RT-PCR和Western blot实验检测相关胶原蛋白表达.结果:ASMq-TF能够显著抑制HFs的增殖及抑制胶原蛋白的表达.讨论:本实验结果表明ASMq-TF通过促进SMAD7合成及减少CollagenⅠ分泌等抑制HFs增殖.%Objective To evaluate in vitro effect of Abnormal savda munziq-total flavonoids on the proliferation and collagen synthesis of human hypertrophic scar fibroblasts(HFs). Methods HFs was cultured in vitro and treated with different concentrations of ASMq-TF.Then,the cell proliferation was detected by CCK-8.The expression of collagen was detected by RT-PCR and Western blot. ResuIts ASMq-TF can inhibit the proliferation of HFs and inhibit the expression of collagen protein Ⅰ. ConcIusion The results of this experiment showed that ASMq-TF inhibited the proliferation of HFs by promoting SMAD7 synthesis and reducing Collagen secretionⅠ.
