低温对沙土鼠脑缺血海马微管运动蛋白Kinesin活性的影响

摘要

目的:观察脑缺血再灌注期间海马锥体细胞微管运动蛋白Kinesin活性的变化及低温对其的影响。方法:沙土鼠前脑缺血再灌注模型,脑缺血时间为10分钟。应用免疫组织化学染色方法结合计算机图像分析技术测定海马微管运动蛋白Kinesin的活性。结果:在海马CA1区,常温缺血再灌注6、48和96小时,微管运动蛋白Kinesin的活性分别降至假手术组的58%、38%和12%(P均<0.01),而低温组在再灌注6、48和96小时后,微管运动蛋白Kinesin活性分别为假手术组的97%、81%和79%,明显高于常温组。在海马CA2、CA3和CA4区,微管运动蛋白Kinesin活性无明显变化。结论:低温可明显抑制脑缺血再灌注期间海马CA1区微管运动蛋白Kinesin活性的下降。%Objective:To investigate the effect of hypothermia on theactivity of Kinesin (a microtubule based motor proteins) in the hippocampus in gerbils during cerebral ischemiareperfusion.Methods:A cerebral ischemia model was reproduced in gerbils by occluding carotid arteries on both sides,and cerebral ischemia lasted for 10 minutes.Kinesin activity in the hippocampus was determined at 6,48 and 96 hours after reperfusion by immunohistochemical staining and examined with a computer image analysis system.Results:In normothermia groups,Kinesin activities in the hippocampus CA1 sector in gerbils significantly decreased at 6,48 and 96 hours after reperfusion,being 58%,38% and 12% of shamoperated controls (all P<0.01).However,Kinesin activities in hypothermia groups were about 97%,81% and 79% of that in shamoperated groups.Hypothermia obviously mitigated the decrement of Kinesin activity in the hippocampus CA1 sector in gerbils.No marked changes in Kinesin activities in the hippocampus CA2,CA3 and CA4 sector were observed during cerebral ischemiareperfusion.Conclusions:Hypothermia can significantly inhibit the decrease in Kinesin activity after cerebral ischemiareperfusion in gerbils.
