利用3-2-2×TopMark杂交组合,得到152株重组自交系群体,构建甜瓜分子遗传图谱.该图谱包括71个SSR标记和94个AFLP标记,由17个连锁群构成,覆盖基因组总长度1222.9 cM,标记之间平均距离为7.41 cM.筛选了428对SSR分子标记(其中360对为EST-SSR引物,68对为g-SSR引物),得到94对具有多态性的SSR标记,其片段大小在150~300 bp之间,多态性比例占21.29%;筛选了256对EcorI/MseI AFLP引物组合,得到22对AFLP多态性引物,共含有146个AFLP多态性位点,其片段大小在100~600 bp之间.SSR标记较平均的分布在染色体上,AFLP标记在LG1、LG12及LG14上出现聚集现象.%Using 152 RILs plants from 3-2-2 × TopMark to construct melon ( Cucumis melo L.) molecular genetic map, a total of 71 SSR markers, 94 AFLP markers were grouped into 17 linkage groups, which spanned 1 222.9 cM with an average marker interval of 7.41 cM.Selected 428 SSR markers including 360 pairs of EST-SSB markers and 68 pairs of g-SSR markers, 94 SSB primers were polymorphic with the size from 150-300 bp; 22 in 256 AFLP EcorI/MseI.146 polymorphic loci with segment size between 100-600 bp were produced.Distribution of SSR markers was evenly on chromosome.AFLP markers were clustered in LG1, LG12 and LG14.
