利用磷盐触发2,4,5-三苯基吡喃盐开环,合成了一种可用于线粒体成像荧光探针的新型的共轭膦盐TPP.该化合物10µmol/L在纯乙腈中几乎不发光,但增加溶度或者增加溶液水含量后,共轭膦盐TPP的荧光强度显著增加,表明该化合物在水溶液中具有优良的聚集诱导荧光增强的性质.通过场发射扫描电镜观察,共轭膦盐TPP能够自组装形成荧光纳米粒子.利用共聚焦显微镜的双通道检测成像实验证实了该化合物具有高度的线粒体选择性.% A new conjugated phosphonium salt TPP, which can be used as fluorescence probes to image mitochondria in cells, has been synthesized readily from phosphine-triggered ring-opening of 2,4,5-triphenylpyrylium salt. When TPP was dissolved in CH3CN to dilute solution (10 μmol/L), almost no fluorescence was observed. Increasing the concentration of the solution or the fractions of water in mixture dramatically enhanced the fluorescent intensity, which indicates the aggregation-induced emission enhancement (AIEE) feature. The shape of nanoparticles was observed by field emission scanning electron microscopy (FE-SEM). Confocal fluorescence images of live HEK-293 cells demonstrate that TPP could yield high intracellular selectivity toward mitochondria.
