大黄素衍生物的合成及灭蚊活性研究

     

摘要

To design and synthesize Emodin derivatives with mosquito larvicidal activity,Emodin as the lead compound was modi-fied with esterification or etherification to ameliorate the solubility and improve the mosquito larvicidal activity.Thirteen Emodin derivatives were prepared and their structures were identified by 1 HNMR and ESIMS.The mosquito larvicidal activity against the fourth-instar larvae of Aedes albopictus was evaluated.The results indicate that the mosquito larvicidal activity decreases when anthraquinone Emodin is reduced to anthrone.Whereas,the etherification or esterification of all the hydroxyl groups in Emodin is helpful to improve the mosquito larvicidal activity.The introduction of Br group at C-6-Me of Emodin is able to enhance the mos-quito larvicidal activity,while the introduction of hydroxy or amino group can increase the activity.%为改善大黄素的生物利用度,提高其灭蚊效果,本文以大黄素为先导化合物,通过酯化或醚化等化学修饰设计合成了系列大黄素衍生物。以大黄素为起始原料合成了13个大黄素衍生物,其化学结构经1 HNMR、ESIMS 等确证,并测定了其对4龄白纹伊蚊幼虫的杀虫活性。构效关系研究表明:将蒽醌大黄素还原成蒽酮后灭蚊活性降低;大黄素被醚化或酯化后,灭蚊活性显著提高;大黄素的 C-6位甲基被溴代后,灭蚊活性下降,而引入羟基或氨基基团后灭蚊活性提高。
