一株降解树脂细菌的鉴定及培养基的优化

         

摘要

采用16SrDNA基因序列比对和进化树分析,鉴定出具有降解天然树脂能力的微生物Z75为enterobacter pulveris.同时,对其进行单因素实验和响应面分析设计实验,得出该菌最优发酵培养基的配方:葡萄糖12.5g/L,酵母粉38.5g/L, MgSO42.5g/L,K2HPO42.5g/L,NaCl 30g/L,pH7.63,接种菌龄OD600=1.300.利用新的培养基配方发酵的发酵液,对反应底物颗粒降解从6.5%提高到22.49%,约提高了3.5倍.%Though 16SrDNA gene sequence alignment and evolutionary tree analysis, it was identified that Z75 which have the ability to degrade natural resin is Enterobacter pulveris. At the same time, single factor choice experiment and response surface analysis design tests on Z75 was carried out, reaching the best formulation of the fermentation medium:glucose 12.5g/l, yeast powder 38.5 g/l, MgSO4 2.5g/l, K2HPO4 2.5g/l, NaCl 30g/l, pH 7.63, inoculant age OD600 is 1.300. Using the fermented liquid of new medium formula, degradation rate increases from 6.5% to 22.49%, increasing by about 3.5 times.
