蟾酥注射液对HL60/ADM细胞多药耐药的逆转作用研究

OBJECTIVE: To investigate the effects of Chansu injection.on reversal of multi-drug resistance (MDR) of HL60/ ADM cell. METHODS: MTT assay was used to investigate the effect of Chansu injection on the proliferation and the adramycin (ADM) sensitivity of HL60/ADM cell. HPLC method was applied to determine the ADM uptake abilities of HL60/ADM cell line. RESULTS: The proliferation of HL60/ADM cell was significantly inhibited by Chansu injection. When pretreated with Chansu injection in noncytotoxic dose, the IC5o of ADM in HL60/ADM cell was significantly reduced(P<0.01) , and ADM uptake abilities of it enhanced significantly (P<0.01). CONCLUSION: Chansu injection can partially reverse the MDR of HL60/ADM cell.%目的:研究蟾酥注射液对人耐阿霉素(ADM)的急性髓性白血病细胞HL60/ADM多药耐药(MDR)的逆转作用.方法:采用MTT法分别考察蟾酥注射液对HL60/ADM细胞增殖能力和对ADM敏感性的变化,并采用高效液相色谱法考察HL60/ADM细胞对ADM的摄取能力.结果:蟾酥注射液能抑制细胞增殖；非细胞毒剂量蟾酥注射液预作用后,ADM对HL60/ADM细胞的半数抑制浓度(IC50)显著降低(P＜0.01),HL60/ADM细胞对ADM摄取能力显著增加(P＜0.01).结论:蟾酥注射液可部分逆转HL60/ADM细胞对ADM的耐药性.
