反相离子对色谱法同时测定黄柏八味片中盐酸黄柏碱和盐酸小檗碱含量

     

摘要

Objective To establish the reversed phase ion-pair chromatography method for the determination of berberine hydrochloride and phellodendrine chloride in Huangbaibawei Tablets. Methods The Agilent C18 column ( 250 mm × 4. 6 mm, 5 μm ) was used. The mo-bile phase was acetonitrile-0. 5% phosphoric acid solution, adjusted to pH 4 with phosphoric acid, each 100 mL added sodium dodecyl sulfate 0. 2 g ( 65∶35 ) . The flow rate was 1. 0 mL/min, the column temperature was 25℃ and the detection wavelength was 284 nm. Results The injection volume of berberine hydrochloride and phellodendrine chloride were in good linear relationship with the peak area in the ranges of 0. 204-1. 020 μg and 0. 506-2. 530 μg . The average recovery rates were 100. 18% and 99. 82%, and the RSD were 0. 14% and 0. 51% ( n=6 ) . Conclusion The method is simple, accurate, with good separation of the 2 components, which can be used as the quality control of Huangbaibawei Tablets.%目的:建立同时测定黄柏八味片中盐酸小檗碱和盐酸黄柏碱含量的反相离子对色谱法。方法色谱柱为Agilent C18柱(250 mm ×4.6 mm,5μm ),流动相为0.5%磷酸溶液(用磷酸调pH至4.0,每100 mL加十二烷基磺酸钠0.2 g)-乙腈(65∶35),柱温25℃,流速1.0 mL/min,检测波长284 nm。结果盐酸小檗碱与盐酸黄柏碱进样量分别在0.204~1.020μg和0.506~2.530μg与峰面积线性关系良好,平均回收率分别为100.18%和99.82%,RSD分别为0.14%和0.51%( n=6)。结论该法简便易行,结果可靠准确,组分分离良好,可用于制剂的质量控制。
