高效液相色谱法测定跌打活血酊中阿魏酸含量

摘要

Objective To establish an HPLC method for the content determination of ferulic acid in Diedahuoxue Tincture by HPLC. Methods The ODS2 C18 ( 150 mm × 4. 6 mm, 5 μm ) was used as chromatographic column;acetonitrile-0. 085% phosphoric acid solution ( 17∶83 ) as mobile phase, the flow rate was 1 mL/min, the detection wavelength was 316 nm, the column temperature was 35 ℃, the injection volume was 10 μL. Results The mass concentration of ferulic acid showed good linear relationship with the peak area in the range of 2. 835-567 μg/mL, the average recovery rate was 99. 04%, RSD was 1. 77%( r=0. 999 8, n=9 );the content of ferulic acid in the 3 batches of samples were 0. 015%, 0. 018%, 0. 012%. Conclusion The established method can quickly determine the content of ferulic acid in Diedahuoxue Tincture, has the advantages of simple operation, accurate result, high sensitivity, and can be used for the quality control of Diedahuoxue Tincture.%目的：建立测定跌打活血酊中阿魏酸含量的高效液相色谱法。方法色谱柱为迪马ODS2 C18柱(150 mm ×4.6 mm，5μm )，流动相为乙腈－0.085%磷酸溶液（17∶83），流速1 mL/min，检测波长316 nm，柱温35℃，进样量10μL。结果阿魏酸质量浓度在2.835~567μg/mL范围内与峰面积线性关系良好（ r=0.9998）；平均加样回收率为99.04%，RSD为1.77%（ n=9）；3批样品中阿魏酸的含量分别为0.015%，0.018%，0.012%。结论该方法可快速测定阿魏酸的含量，操作简便、结果准确、灵敏度高，可用于跌打活血酊的质量控制。
