板蓝根饮片、板蓝根颗粒中(R,S)-告依春的含量测定

         

摘要

目的 建立测定板蓝根饮片与颗粒中(R,S)-告依春含量的高效液相色谱(HPLC)法.方法 采用超声提取进行样品处理,以HPLC法测定指标成分(R,S)-告依春的含量.色谱柱采用Agilent C18柱(250 mm×4.6 mm,5μm),柱温为35℃,检测波长为245 nm,流动相为甲醇-水(15:85),流速为1.0 mL/min.结果 用水作为溶剂超声(功率250 W,频率30 kHz)1 h为提取方法较煎煮法高效、环保;(R,S)-告依春质量浓度在0.0031~0.0434 g/L范围内与峰面积线性关系良好(r=0.99994);饮片和颗粒中(R,S)-告依春平均回收率分别为98.08%和97.86%,RSD分别为0.79%和0.55%(n=6).结论 该方法易于掌握、准确度高、重复性好,适用于板蓝根饮片、板蓝根颗粒中指标成分(R,S)-告依春的含量测定.%Objective To establish an HPLC method for content determination of ( R, S ) -epigoitrin in Banangen Granules and Radix isatidis. Methods The sample was processed by ultrasonic extraction, the content of ( R, S ) -epigoitrin was determined by HPLC method. Agilent C18 column ( 250 mm × 4. 6 mm, 5 μm ) was adopted, the column temperature was 35 ℃, the detection wavelength was 245 nm, the mobile phase was methanol-water ( 15 :85 ) , and the flow rate was 1. 0 mL/min. Results Ultrasonic extraction method by using water as solvent to extract in 1 h ( power 250 W, frequency 30 kHz ) was more environment-friendly and high efficient than the method of decoction extraction, and ( R, S)-epigoitrin had good linear relationship in the range of 0. 0031-0. 0434 g/L( r=0. 99994). The average recovery rates of ( R, S ) -epigoitrin in Banangen Granules and Radix isatidis were 98. 08% and 97. 86%, and RSDs were 0. 79% and 0. 55% ( n=6 ) . Conclusion The method is easy to grasp, it has high accuracy and good reproducibility, which is suitable for the content determination of ( R, S ) -epigoitrin in Banangen Granules and Radix isatidis.
