兔骨缺损ESW治疗后成骨细胞中VEGF的表达

         

摘要

目的:蔑察体外冲击渡(ESW)干预兔骨缺损模型后成骨细胞中血管内皮细胞生长因子(VEGF)的表达情况,初步探讨其促进成骨作用的分子生物学机制.方法:将18只中国健康家兔制备双桡骨骨缺损动物模型,右上肢作为实验组,左上肢为对照组,1周后全麻下行右上肢ESW治疗,左上肢不做治疗.定期拍X线片、组织学观察、免疫组化检测.结果:①X线光片显示冲击波治疗4、6周后,治疗组骨愈合较对照组好;②组织学观察显示治疗组骨细胞、成纤维细胞、毛细血管内皮细胞、成骨细胞、间充质细胞等细胞增生活跃;③免疫组化方法示治疗组成骨细胞中YEGF含量明显高于对照组,差异有统计学意义(P<0.05).结论:ESW干预后成骨细胞中生物学活性因子VEGF表达增加,可能是EsW促进成骨细胞成骨活动的有力证据.%Objective:To investigate ESWT's influences on the expression of VEGF in bonycallus,and the further effect of ESWT's mechanism about how to cure bone defect.Methods:IS Chinese healthy domestic rabbits were made the animal models of the double radial bone defection and damage,right upper extremity was experiment set,left upper extremity was matched control,after the wound healed about a week,right upper extremity enforce ESWT under general anesthesia,carry to correspond part at the bone defection and damage two pieces,the left opper extremity did't do it. Then we took X-ray photo to observe condition of bone defection after 4 weeks and 6 weeks. And taking the bone defection' s bony callus organization specimen; we would do histology-checking to observe organization propagate of cell circumstance,and do immunohistochem-istry -checking to observe expression of VEGF.Results:The content of VEGF in the osteoblast of the Extracorporeal shock wave treatment set be more obvious than the matched team,bone cell,fibroblast,blood capillary,osteoblast, mesenehymal of ESWT set increased actively.Conclusion:The VEGF's expression can be boosted under the ESW's stimulation, make the cell propagate to divide and help the bone defect to heal.
