益气活血解毒方治疗晚期卵巢癌有效可能的单核苷酸多态性位点及基因靶点

摘要

目的 探究益气活血解毒方治疗晚期卵巢癌有效可能的单核苷酸多态性(SNPs)位点及基因靶点.方法 选取2015年3月至2016年12月就诊于中国中医科学院广安门医院的气虚血瘀型晚期卵巢癌患者300例.服用益气活血解毒方半年后抽取外周血,按照中位无进展生存期分为2组,通过2组的对比筛选出显著差异的SNPs位点及可能的对中药敏感的基因靶点.结果 筛选出差异的SNPs位点共计242 901个,其中存在显著差异的变异位点1 773个.筛选出信使RNA的差异表达基因158个,其中上调基因58个,下调基因100个.将筛选出的SNPs位点进行基因定位并与差异表达基因取交集,得到与中药干预疗效差异有关的基因为:CD40、AFF3、EIF2AK3、WNK1、FYB、FBXL13、PLXNC1、BCL11A、IFI16、GNG7.结论 中药治疗晚期卵巢癌的有效性可能与SNPs位点及基因靶点有关.%Objective To explore the possible effective single nucleotide polymorphism (SNPs) sites and gene targets of Yiqi Huoxue Jiedu decoction in the treatment of advanced ovarian cancer.Methods Three hundred patients with qi-deficiency and blood-stasis type advanced ovarian cancer who visited Guang'anmen Hospital,China Academy of Traditional Chinese Medicine from March 2015 to December 2016 were enrolled.All patients were treated with Yiqi Huoxue Jiedn decoction;peripheral blood was taken 6 months after treatment for gene test.The patients were divided into 2 groups according to the median progression-free survival time;SNPs sites and gene targets sensitive to Chinese herbal treatment were analyzed.Results A total of 242 901 SNPs were screened out,of which 1 773 sites showed significant differences.Differentially expressed genes of messenger RNA were 158 sites,including 58 up-regulated genes and 100 up-regulated genes.Genes involved in the differences of therapeutic efficacy of Chinese herbs were CD40,AFF3,EIF2AK3,WNK1,FYB,FBXL13,PLXNC1,BCL11A,IFI16 and GNG7.Conclusion Effectiveness of traditional Chinese medicine in the treatment of advanced ovarian cancer may be related to SNPs and gene targets.
