降浊颗粒对慢性肾衰竭大鼠肾组织纤维连接蛋白的影响

     

摘要

目的 本研究拟通过动物实验,观察降浊颗粒对5/6肾切除慢性肾衰竭(CRF)大鼠肾组织纤维连接蛋白(FN)的影响,来探讨降浊颗粒对CRF的治疗效果及抗肾间质纤维化的机制.方法 选择健康Wistar雄性大鼠120只,随机分为假手术组及模型组.5/6肾大部切除法建立CRF大鼠模型.模型组大鼠随机分为五组给药.动物灌胃给予生理盐水或给药,连续60 d.观察实验大鼠治疗前后的体征及一般状态,血清肌酐(Scr)、尿素氮(BUN)的变化,取大鼠肾脏病理切片.采用免疫组织化学染色(SP法)检测FN表达情况.结果 降浊颗粒能够降低CRF大鼠血清BUN、Scr水平,贝那普利组、降浊颗粒高、中、低剂量组与模型对照组比较差异有统计学意义(P<0.05),高、中、低剂量组与贝那普利组比较,差异无统计学意义(P>0.05);降浊颗粒能够降低实验大鼠肾组织中的FN的表达,降浊颗粒高剂量组与模型对照组比较差异有高度统计学意义(P<0.01),降浊颗粒高、中剂量组与贝那普利组比较差异无统计学意义(P>0.05).结论 降浊颗粒能够减少CRF大鼠血清BUN、Scr水平,抑制大鼠肾组织FN的表达,干预肾间质纤维化.最终延缓大鼠CRF进展,以降浊颗粒高、中剂量组效果最好,其疗效与贝那普利相当.%Objective To observe the therapeutic effects of Turbidity-reducing Granule on kidney tissues fibronectin (FN) of rats with chronic renal failure (CRF), so as to observe the mechanism of the granule in the treatment of CRF and renal Interstitial fibrosis. Methods 120 healthy male Wistar rats were selected and randomly divided Into sham operation group and model group, 5/6 subtotal nephrectomy method was used to establish CFR rats. Rats in model group were randomly divided into 5 groups for administration. Rats were gastric-ally administered Turbidity-reducing Granule or saline for 60 consecutive days. The rats were observed before and after treatment of signs and general condition, serum BUN, Scr changes. Immunohistochemical staining (SP method} were used to detect FN expression. Results Turbidity reducing Granule can reduce the level of serum BUN, Scr in rats with CRF, there were statistically significant differences of Benalapril group and high, medium, low dose of Turbidity-reducing Granule group compared with model group (P < 0.05), while there were no significant differences of high, medium, low dose of Turbidity-reducing Granule group compared with Benalapril group (P > 0.05). Turbidity-reducing Granule downreguiated FN in the experimental rat kidney tissue In high-dose group, and had significant difference with model control group (P < 0.01), there was no significant difference in high, medium dose treatment group and Benalapril group (P > 0.05). Conclusion Turbidity-reducing Granule can reduce the level of BUN ,Scr of CRF rats, reduce CRF inhibit rats kidney tissues FN expression, intervene renal interstitial fibrosis, delay the progress of CRF in rats, especially for the the group of high, medium dose group, the efficacy Is similar to Benalapril.
