舒筋健腰丸对坐骨神经痛大鼠坐骨神经组织及血清细胞因子的影响

摘要

Objective To study the effect of Shujin Jianyao Pills on the pathological changes of ischiadic nerve and cytokine in the serum of CCI (chronic constriction injury) rats.Methods 112 SD rats were randomly divided into sham operation group,model group,Zhuanggu Guanjie Pill group,Paracetamol and Caffeine Tablets group,Shujin Jianyao Pill low dose group (1.10 g/kg),middle dose group (2.19 g/kg) and high dose group (4.38 g/kg),with 16 rats in each group.The right common sciatic nerve was exposed by blunt dissection,and four ligatures were tied loosely around the sciatic nerve.Sham operation referred to the procedure of exposing the nerve as above but without any nerve ligation.All groups were administrated with corresponding drugs by intragastric administration once a day for 28 days.After the last administration,the hindpaw withdrawal latency (HWL) of the rats,the contents of inflammatory cytokines including TNF-α,IL-1β,IL-6,PGE2 in the serum and the pathological changes of ischiadic nerve were detected.Results Compared to the sham operation group,in the model group,the HWL was shortened,ischiadic nerve was becomne swollen and vacuolar,the serum levels of IL-1β and IL-6 were increased (P ＜ 0.05).Compared to the model group,in the Shujin Jianyao Pill groups,the HWL were extended,the pathological changes were ameliorated induced by CCI,and the IL-1β and IL-6 levels were decreased (P ＜ 0.05).Conclusion Shujin Jianyao Pill can alleviate the histologic changes of sciatic nerve,inhibit the expression of inflammatory cytokines,which has therapeutic effect on traumatic neuralgia.%目的 研究舒筋健腰丸对坐骨神经痛大鼠的坐骨神经组织病理改变以及血清中细胞因子的影响.方法 选取112只SD大鼠,随机分为假手术组,模型组,壮骨关节丸组,芬必得酚咖片组,舒筋健腰丸高(4.38 g/kg)、中(2.19 g/kg)、低(1.10 g/kg)剂量组,每组16只.通过钝性分离,暴露大鼠右下肢坐骨神经并对其进行4道结扎,制备坐骨神经痛模型.假手术组只暴露坐骨神经但不结扎坐骨神经.造模成功后,连续灌胃给药28 d.末次给药后,检测各组大鼠的缩爪潜伏期,血清中TNF-α、IL-1β、IL-6、PGE2的含量及坐骨神经组织的病理学改变.结果 与假手术组相比,模型组大鼠缩爪潜伏期明显缩短(P＜0.05),坐骨神经发生水肿和空泡变性,血清中IL-Iβ、IL-6含量显著升高(P＜0.05).与模型组相比,舒筋健腰丸组缩爪潜伏期延长(P＜0.05),坐骨神经病的变程度减轻,血清中炎性因子IL-1β、IL-6含量下降(P＜0.05).结论 舒筋健腰丸能明显减轻坐骨神经组织学改变,抑制炎性因子表达,对损伤性神经痛具有治疗作用.
