白介素-10对去卵巢小鼠骨质疏松的作用

         

摘要

目的 探讨白介素-10(IL-10)对去卵巢小鼠骨质疏松的影响.方法 采用随机数表法将60只ICR小鼠随机分为对照组、模型组(OVX组)和治疗组(IL-10组),每组20只.OVX组小鼠切除双侧卵巢复制骨质疏松动物模型;IL-10组小鼠切除双侧卵巢后尾静脉注射IL-10(80 mg/kg),2次/周,连续给药12周;对照组只切除卵巢周围与卵巢大小相当的一块脂肪组织.12周后检测各组小鼠骨相关参数以及血清骨源性碱性磷酸酶(BALP)、白介素-1β(IL-1β)、白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)含量.结果 OVX组小鼠造模后,骨密度(BMD)、骨体积分数(BWTV)、骨小梁数量(Tb.N)和骨小梁厚度(Tb.Th)明显下降,骨小梁分离度(Tb.Sp)显著上升,与对照组比较,差异有高度统计学意义(P<0.01).OVX组小鼠呈现明显的骨质疏松表现.而与OVX组比较,IL-10治疗组小鼠BMD、BV/TV、Tb.N和Tb.Th明显上升,Tb.Sp明显下降,差异有统计学意义(P< 0.05或P< 0.01).Micro-CT分析图像显示,OVX组小鼠骨小梁显著减少,而IL-10治疗组小鼠股骨远端骨小梁减少程度有明显改善.与对照组比较,OVX组小鼠血清BALP、IL-1β、IL-6和TNF-α水平明显增高,差异有高度统计学意义(P<0.01);而同时给予IL-10治疗后,小鼠血清BALP、IL-1β、IL-6和TNF-α水平明显下降,与OVX组比较,差异有统计学意义(P<0.05或P<0.01).结论 IL-10能够增加去卵巢骨质疏松症小鼠BMD,降低BALP、IL-1β、IL-6和TNF-α水平,减轻骨质疏松症状.IL-10在去卵巢小鼠骨质疏松的治疗上有明显作用.%Objective To explore the effect of interleukin-10 (IL-10) on osteoporosis in ovariectomized mice.Methods 60 ICR mice were divided into three groups:control group,model group (OVX group) and treatment group (IL-10 group) by using random data table,each group had 20 mice.Bilateral ovaries in mice of OVX group were removed to establish ovariectomized mice model.Bilateral ovaries in mice of IL-10 group were removed,followed by IL-10 injection (80 mg/kg) for 12 weeks in a row (2 times per week).Only a piece of fat tissue size same as ovary around the ovary was removed in the control group.After 12 weeks,bone related parameters,bone alkaline phosphatase (BALP),IL-1β,IL-6 and tumor necrosis factor (TNF)-α of the bone tissue were measured.Results Compared with control group,bone mineral density (BMD),bone volume fraction (BV/TV),trabecular thickness (Tb.Th) and trabecular number (Tb.N) significantly decreased,and trabecular separation (Tb.Sp) increased in OVX group,with statistically significant differences (P < 0.01).The OVX group animals had obvious performance of osteoporosis.After IL-10 treatment,compared with OVX group,BMD,BV/TV,Tb.Th and Tb.N significantly increased,and Tb.Sp decreased,with statistically significant differences (P < 0.05 or P < 0.01).Micro-CT images showed that trabecular number in OVX group decreased obviously,and increased significantly in IL-10 group.Compared with control group,serum BALP,IL-1β,IL-6 and TNF-α significantly increased in OVX group (P < 0.01);after IL-10 treatment,serum BALP,IL-1β,IL-6 and TNF-α decreased obviously in IL-10 group,compared with OVX group (P < 0.05 or P < 0.01).Conclusion IL-10 can alleviate the symptoms of osteoporosis,increase the BMD and decrease levels of BALP,IL-1β,IL-6 and TNF-α in serum.IL-10 has an apparent role of antiosteoporosis in ovariectomized mice.
