川芎嗪注射液对缺氧状态下视网膜血管内皮细胞增殖作用的影响

         

摘要

目的 观察川芎嗪注射液对缺氧状态下视网膜血管内皮细胞生长、增殖的影响.方法 体外培养牛视网膜血管内皮细胞( RVEC),将传代的细胞分成3组,即正常细胞对照组(DMEM常规培养基培养)、缺氧模型组、缺氧+不同浓度川芎嗪组(含0.002、0.02、0.2和2 μg/ml川芎嗪的培养基).培养24、48 h后在倒置相差显微镜下观察细胞形态学改变;四甲基偶氮唑盐(MTT)法检测各组血管内皮细胞的增殖情况.结果 缺氧组及各药物组各时间点的光密度值明显低于正常细胞对照组(P<0.01);川芎嗪0.002 μg/ml组的光密度值与缺氧组比较无统计学差异(P>0.05),0.02、0.2 μg/ml组的光密度值与缺氧组比较有统计学差异(P<0.01),其中0.2 μg/ml组的数值最高,但仍低于正常细胞对照组;而川芎嗪2 μg/ml组的光密度值有下降趋势.结论 川芎嗪可抑制缺氧引起的视网膜血管内皮细胞活性降低,对细胞的增殖有一定的促进作用.%OBJECTIVE To observe the effect of ligustrazine injection on growth and prolif-eration of retinal vascular endothelial cells in hypoxia. METHODS In vitro bovine retinal vascular endothelial cells (RVEC), were divided into three groups, namely the normal cells in the control group (routine DMEM medium), hypoxia model group, the concentration of oxygen + different ligustrazine groups (0.002, 0.02, 0.2 and 2 μg /ml) of ligustrazine medium. The morphological changes of cells morphology were observed under inverted phase contrast microscope after 24 h and 48 h. The proliferation of vascular endothelial cells in each group was examined by MTT. RESULTS The optical density value of each time point was significantly lower than that in the control group (P<0.01). There was no statistically significant difference (P>0.05) between 0.002 μg/ml Ligustrazine group and hypoxia group, but statistically significant difference (P<0.01) in 0.02, 0.2 μg/ml group, of which 0.2 μg/ml group had the highest value, but they were still lower than the normal cells in the control group. The optical density of 2 μg/ml ligustrazine group was decreased. CONCLUSIONS Ligustrazine could inhibit the activity of retinal vascular endothelial cells induced by hypoxia and promote the proliferation of the cells.
