糠醛渣酶解工艺优化及过程的研究

     

摘要

Based on the data of the single factor experiment, respond surface methodology (RSM) was utilized to optimize the conditions for enzymatic hydrolysis of furfural residue pretreated by overliming. Furfural residue was hydrolyzed in 5Lfermentor, and the data of different phases were analyzed simultaneously. The results showed that the optimized process conditions for the residue enzymatic hydrolysis were: 46.8℃ of reaction temperature, 4.32 of pH, 35.95 FPU/g of cellulose, and 2.899% of substrate concentration, the average conversion rate could reach 92.59%. The simultaneous analysis indicated that enzymatic hydrolysis reacted quickly in the early period: the saccharification rate reached 55.33% and 87.88% at 3h and 18h from reaction beginning, respectively. The highest saccharification rate was up to 98.1% at 36h, which was much shorter than other cellulosic materials.%笔者以糠醛渣为原料,在单因素实验的基础上,应用响应曲面法进行酶解糖化试验,探索了纤维素酶水解反应的最佳工艺条件.在优化条件的基础上进行了发酵罐糖化试验,同步分析了反应过程中糖化率的变化.研究结果表明:糠醛淹酶解的最佳工艺条件为:温度46.8℃,pH 4.32,酶用量35.95FPU/g,底物浓度2.899%,纤维素的平均转化率为92.59%.糖化过程同步分析结果显示,反应初期糖化速度非常快,反应进行3h糖化率达到了55.33%.反应18h时糖化率已达到87.88%,36h酶解反应基本结束,糖化率高达98.1%,比其他纤维原料的糖化时间大大缩短.
