七叶亭对大鼠血管平滑肌细胞凋亡的调节作用及分子机制研究

         

摘要

目的:研究七叶亭对大鼠血管平滑肌细胞凋亡的调节作用及分子机制。方法离体培养大鼠血管平滑肌细胞,分为高剂量组、中剂量组、低剂量组和对照组。采用MTT法检测细胞的凋亡情况,采用荧光定量PCR检测细胞中Bcl-2、Bax和caspases-9的mRNA含量。结果①细胞凋亡:七叶亭处理后MTT值均低于对照组,且药物剂量越高、MTT值越低;②基因mRNA含量:七叶亭处理后,Bcl-2的mRNA含量低于对照组,Bax和caspases-9的mRNA含量高于对照组;且药物剂量越高,Bcl-2的mRNA含量越低,Bax和caspases-9的mRNA含量越高。结论七叶亭处理能够诱导大鼠血管平滑肌细胞发生凋亡,且抑制Bcl-2表达和促进Bax、caspases-9表达可能是药物发挥作用的分子机制。%Objective To study the regulation effect and molecular mechanisms of esculetin on rat vascular smooth muscle cell apoptosis. Methods Rats' vascular smooth muscle cells were cultured and divided into high-dose group, middle-dose group, low-dose group and control group. Cell apoptosis was detected by MTT assay. Bcl-2, Bax and caspases-9 mRNA contents were detect-ed by quantitative PCR. Results ①Cell apoptosis: after esculetin treatment, MTT values were lower than control group and the higher the dose, the lower the MTT value;②mRNA content:after esculetin treatment, Bcl-2's mRNA levels were lower than con-trol group, Bax and caspases-9 mRNA levels were higher than control group; the higher the dose, the lower the mRNA levels of Bcl-2 and the higher the mRNA levels of Bax and caspases-9. Conclusion Esculetin treatment can induce the apoptosis of rat vascular smooth muscle cells and inhibit the Bcl-2 expression and promote Bax and caspases-9 expression, which may play a role in the molecular mechanisms of drugs.
