二氧化碳用于Nisin浓缩分离的研究

         

摘要

泡沫分离是浓缩分离乳酸链球菌素常用的方法,本文研究了一种利用二氧化碳的泡沫分离方法来提高收率、降低残液效价、缩短分离时间.针对装液量、气速、pH及起泡性能等几个影响泡沫分离效果的操作条件进行研究,使用二氧化碳均能获得相对较好的分离效果,说明二氧化碳应用到Nisin发酵液浓缩分离具有可行性,在800mL的装液量、气速100mL/min、pH2.5的条件下二氧化碳泡沫分离的收率达到92%比空气泡沫分离高14%,富集比提高1.5,分离时间缩短1h.间隔15min通入2~3min二氧化碳的操作方式,其收率达到90.3%,富集比达到7.5,并能缩短分离时间,说明间歇式二氧化碳泡沫分离是控制分离成本提高分离效果比较理想的操作策略.%Foam fraction is a common technique used for precipitating Nisin from fermentation, In this article, we found a new method for increasing recovery, reducing residual liquid potency, shorting the separation time is to add CO2 to process. According to the results of operating conditions which affect the foam fraction, such as liquid volume, gas flow rate, pH and foaming properties etc, we found that the foam separation effect of C02 is always better than the air, which shows the feasibility of carbon dioxide applied to the Nisin fermentation concentration and separation. In the circumstances of 800mL of liquid volume, lOOmL/min of gas flow rate and pH 2. 5, the foam separation of carbon dioxide recovery ratio is 92% and 14% higher than the air separation, the enrichment ration increased 1. 5 and the separation time is shorten one hour. Adapting the way of pulsing 2 ~ 3min CO2 every 15mins, the recovery ratio reached 90. 3% , the enrichment reached 7. 5 and shorten the separation time which shows that batch foam separation by pulsing carbon dioxide is the ideal operating strategy of control separation cost and improve recovery.
