核壳型单磷酸腺苷分子印迹聚合物的制备

     

摘要

Using polystyrene microsphere (CP)as a core,adenosine-5′-monophosphate(AMP)as a template, acrylamide(AM)as a functional monomer and ethylene glycol dimethacrylate(EGDMA)as a cross-linker,while dimethylsulfoxide(DMSO)was chosen as a porogen and azobisisobutyronitrile(AIBN)as an initiator,core-shell adenosine-5′-monophosphate molecular imprinted polymer(AMP-MIP)was prepared.The effects of different e-lution methods,solvents and polymerization time on the properties of AMP-MIP recognition were discussed. The morphology and property of AMP-MIP were characterized by SEM,the suspension in aqueous medium and the binding experiments.The optimal process conditions were obtained as follows:the elution method was ultra-sound,the elution solvent was methanol-acetic acid (4∶1 )and polymerization time was 30 min.AMP-MIP showed good selectivities not only for AMP but also for glyceraldehyde-3-phosphate dehydrogenase (gapA) which was ADP-ribosylated protein.The adsorption capacity of gapA could reach 3.9 μg·mg-1 .%以聚苯乙烯微球(CP)为内核、单磷酸腺苷(AMP)为模板、丙烯酰胺(AM)为功能单体、乙二醇二甲基丙烯酸酯(EGDMA)为交联剂,制备了核壳型单磷酸腺苷分子印迹聚合物(AMP-MIP),讨论了不同洗脱方式、洗脱溶剂和聚合时间对 AMP-MIP 吸附性能的影响,通过扫描电镜、水中悬浮实验和吸附实验对 AMP-MIP 的形貌和性能进行了表征,得到最佳工艺条件:洗脱方式为超声、洗脱溶剂为甲醇-乙酸(4∶1,体积比)、聚合时间为30 min。分子印迹聚合物不仅对小分子模板有吸附作用,而且对大分子 ADP-核糖基化的甘油醛-3-磷酸脱氢酶(gapA)具有识别作用,吸附量可达3.9μg ·mg-1。
