4NQO诱导小鼠舌癌前病变模型的研究

             

摘要

Objective To establish an animal model of tongue precancerous lesions by using the carcinogen 4-nitroquinoline-1-oxide (4NQO). Methods Thirty C57BL/6J mice were divided into 3 groups,and given 50μg/ml 4NQO as drinking water for 8,12,16 weeks. Sixteen mice served as controls. The mice were sacrificed on weeks 8,12 and 16. All the mice were injected with 5-bromodeoxyuridine ( BrdU) before they were sacrificed. Macroscopic and histopathological observations were performed on the samples of the tongue. Bromodeoxyuridine staining of the tissue was examined by immunohistochemistry. Results Macroscopic observation showed that white patches appeared in the tongue of the groups after 8-16 weeks of treatment by 4NQO. The numbers of mild and moderate dysplasia in the 4NQO-treated groups were 0. 20 ±0. 45,1. 60 ± 1. 14,2. 60 ±0. 82 respectively. The number of BrdU-labeled cells was more in the 4NQO-treated groups than in the controls. Conclusion A mouse model of tongue precancerous lesions can be established by 50μg/ml 4NQO as drinking water.%目的 使用4NQO诱导并建立C57BL/6J小鼠舌癌前病变模型.方法 46只6~8周龄C57BL/6J小鼠,随机取16只作为阴性对照,不做任何处理,分别于8、12、16周末处死.实验组30只饮用50μg/ml 4NQO水溶液,分别于8、12、16周末处死.处死前2小时腹腔注射BrdU (50mg/kg),肉眼及组织病理学观察病变情况,免疫组织化学检测BrdU染色,并进行统计学分析.结果 实验组8、12、16周小鼠,肉眼观察舌背黏膜逐渐粗糙发白,呈白斑样改变;HE染色显示,实验组8、12、16周轻中度异常增生灶数分别为0.20±0.45、1.60±1.14、2.60±0.82;随着病变程度的加重BrdU染色阳性细胞增加,与阴性对照组比较有显著差异.结论 50μg/ml4NQO水溶液8~16周可以诱发小鼠舌癌前病变.
