烟碱型乙酰胆碱受体α3-nAChR和α7-nAChR在口腔癌中的表达

摘要

目的：研究烟碱型乙酰胆碱受体α3-nAChR和α7-nAChR在口腔鳞状细胞癌( oral squamous cell carcinoma, OSCC)中的表达,为口腔癌发病机制的研究提供实验依据。方法随机选取20例福尔马林固定的口腔鳞状细胞癌组织标本,10例正常口腔黏膜作为对照。采用免疫组织化学染色和荧光定量PCR方法,检测烟碱型乙酰胆碱受体α3-nAChR和α7-nAChR在OSCC中的表达。结果口腔鳞状细胞癌中α3-nAChR的mRNA水平是正常口腔黏膜的3.97倍( P <0.05),蛋白表达水平显著高于正常口腔黏膜( P <0.05),α7-nAChR 的mRNA水平是正常口腔黏膜的8.41倍( P<0.05),蛋白表达水平也显著高于正常口腔黏膜( P<0.05)。结论α3-nAChR、α7-nAChR与烟草相关口腔鳞状细胞癌的发生密切相关,在 OSCC治疗中有望成为有价值的分子靶点。%Objective To investigate the expression ofα3-nAChR andα7-nAChR in oral squamous cell carcinoma ( OSCC) and the pathogenesis of tobacco related OSCC. Methods Twenty cases of formalin fixed OSCC tissue samples were randomly selected, and ten cases of normal oral mucosa tissues served as control. The expression of α3-nAChR andα7-nAChR was examined by immunohistochemistry and real-time quantitative PCR. Results Compared with normal oral mucosa,α3-nAChR and α7-nAChR mRNA level and protein expression were increased in oral squamous cell carcinoma ( P<0. 05 ) . Conclusion α3-nAChR and α7-nAChR may be closely related to the occurrence of tobacco-related oral squamous cell carcinoma, and hopefully become therapeutic targets in oral squamous cell carcinoma.
