大鼠实验性蛛网膜下腔出血后脑内HSP72表达增加

     

摘要

目的 研究大鼠蛛网膜下腔出血后HSP72在大脑皮质和海马区的表达.方法 36只Wistar大鼠随机分为假手术组(SO)、枕大池1次注血组(SI)及2次注血组(DI)分别制作蛛网膜下腔出血模型.制备模型成功后取脑行免疫组化染色及Western blot检测海马和大脑皮质区HSP72的表达.结果 海马及大脑皮质有HSP72的表达;与假手术组相比,注血组海马及大脑皮质区HSP72的表达显著增加(SI:0.63±0.17,0.62±0.25;DI:0.81±0.25,0.83±0.34;P<0.05).结论 蛛网膜下腔出血可造成大鼠早期脑损伤,并引起海马及脑皮质HSP72的过度表达.%Objective To assess the expression of HSP72 in cortex and hippocampus after experimental subarachnoid hemorrhage in rats.Methods Thirty-six wistar rats were randomly divided into three groups: sham operation,single intracisternal blood injection and double intracisternal blood injection.The expression of HSP72 in hippocampus and cerebral cortex was detected by immunohistochemistry and Westernblot technique.Results HSP72 expression was detected in intracisternal blood injection group by immunohistochemistry.Semiquantitative analysis of HSP72 electrophoretic bands indicated more expression in hippocampus and cerebral cortex in both injection groups compared with the sham group(SI: 0.63 ±0.17, 0.62 ±0.25; DI: 0.81 ±0.25, 0.83 ±0.34; P <0.05).Conclusion Subarachnoid hemorrhage produced by single and double intracisternal autogenious blood injection can cause HSP72 overexpression in hippocampus and cerebral cortex.
