应用组织芯片技术研究胃癌组织中Mcl-1蛋白的表达及其意义

         

摘要

Objective To study the expression of Mcl-1 in gastric cancer and its clinical significance. Methods Tissue microarrays containing gastric cancer tissue (n = 148) and gastric normal tissue (n =20) were prepared and the expression of Mcl-1 was detected by immunohistochemistry. Results Immunohistochemistry showed that gastric cancer tissues and gastric normal tissue both had the expression of Mcl-1 protein, located in the cytoplasm, pale yellow to dark brown color. In gastric cancer tissues and gastric normal tissue Mcl-1 protein expression rates were 83.78% (124/148) , 60.00% (12/20) , respectively. The difference was statistically significant (x2 = 6.464, P =0.011). In gastric cancer tissues, the expression of Mcl-1 protein with lower degree of tumor differentiation was enhanced, and was associated with lymph node metastasis, but not associated with the patient's age, sex, tumor size, and the tumor infiltration depth (P > 0.05). Conclusion The Mcl-1 protein expression of the gastric cancer tissues is heightened, which is closely related with the development of gastric cancer,its differentiation degree, lymph node metastasis.%目的 探讨胃癌组织髓样细胞白血病蛋白-1(Mcl-1)蛋白的表达与临床病理特征的相关性及其与胃癌发生、发展的关系.方法 用组织芯片和免疫组化的方法 检测148例胃癌组织、20例胃正常组织(含正常胃黏膜组织及癌旁正常胃组织)中Mcl-1蛋白的表达.结果 胃癌组织、胃正常组织中均有Mcl-1蛋白的表达,定位于细胞浆中,着色呈淡黄色至深棕色;胃癌组织、胃正常组织Mcl-1蛋白阳性表达率分别为83.78%(124/148)、60.00%(12/20),差异具有统计学意义(χ2=6.464,P=0.011).胃癌组织中Mcl-1蛋白的表达随肿瘤分化程度的降低而增强,与是否伴有淋巴结转移相关;但与患者的年龄、性别、肿瘤大小、浸润深度等无关.结论 胃癌组织中Mcl-1蛋白的表达增高,与胃癌的发生、分化程度、淋巴结转移密切相关.
